| Objective To observe the impact of ovarian transplantation of castrated rats on thechanging number of hippocampal neurons and the expression of G protein-coupled receptor30(GPR30), postsynaptic density95(PSD-95) and Neurogranin (Ng) to evaluate ovariantransplantation on the protective effect in hippocampal neurons.Method The adult female SD rats (body weight200±20g) were randomly divided intonormal control group, ovariectomized group (OVX group) and ovarian transplant group. Ratsof OVX group and ovarian transplant group were castrated (OVX)in anoestrum. Rats oftransplantation group were transplanted the ovaries of SD rats within3days under the renalcapsule in the7days after castration. The rats in each group were sacrificed in7d,14d,21d and28d. To count the number of hippocampal neurons and observe neuronal morphologyby Nissl staining and observed expression of GPR30, PSD-95and NG byimmunohistochemical method and measure expression of GPR30and PSD-95by Western-blot.Ovarian transplant group was compared with the OVX group and normal control group in thesame period.Results (1) morphology showed that: morphological injury of hippocampal neuronsgradually worsened and the number hippocampal neurons have gradually reduced in the OVXgroup with the castration time extending, compared with the normal control group, thedifference was statistically significant (P<0.05); with the migration time extending,hippocampal neuronal morphology in ovarian transplant group gradually returning to normaland the number of hippocampal neurons have gradually increased, compared with the normal control group, except CA1region28d group, the difference was statistically significant(P<0.05).(2) Immunohistochemical staining showed that: the expression of GPR30, PSD-95andNG in the OVX group have gradually reduced with the castration time extending, comparedwith the normal control group, the difference was statistically significant (P<0.05); theexpression of GPR30, PSD-95and NG in ovarian transplant group have gradually increasedwith the migration time extending, compared with the OVX group in the same period, thedifference was statistically significant(P<0.05), compared with the normal control group,except GPR30’s28d group, NG’s DG region28d group, the remaining group’s differencewas statistically significant (P <0.05).(3) western-blot results of observation: GPR30andPSD-95expression are similar with immunohistochemical results. The expression of GPR30and PSD-95in the OVX group have gradually reduced with the castration time extending,compared with the normal control group, the difference was statistically significant (P<0.05);the expression of GPR30and PSD-95in ovarian transplant group have gradually increasedwith the migration time extending, compared with the normal control group, except GPR30’s21d and28d group, the remaining group’s difference was statistically significant (P <0.05).Conclusion (1) Ovariectomized rat hippocampal neurons arrange sparsely, the numberof layers and the quantity of neurons reduce. The expression levels of GPR30, PSD-95andNG decline;(2) after ovarian transplantation, the number of impaired hippocampal neurons canrecover and neurons arrange in neat rows, the expression of GPR30, PSD-95and NGgradually increased;(3) Ovarian hormone can play a protective effect on the hippocampalneurons through improving the expression of related neurotrophic factor. |
