PART ONEEffect of HBV on Expressions of MIA2in Hepatoma CellsObjective To investigate the relationship of the MIA2expression in hepatoma cells withHBV infection status.Methods Cultured HBV-negative and HBV-positive hepatoma cell lines HepG2andHepG2.2.15cells,to investigate MIA2protein expression differences in two cell lines byimmunohistochemistry and Western blot analysis.Results The MIA2protein expression level reduced in the HepG2.2.15cell linecompared with HepG2cell lines, the results are statistically different.Conclusion MIA2protein expression in HBV-positive liver cancer cells reduced.Theresults indicat a possibility that HBV infection may lead to liver cancer development throughinhibiting MIA2expression. PART TWOEffect of5-AZA-2-deoxycytidine on the expression of MIA2inHepatoma CellsObjective this study was to observe the effet of methylase inhibitor5-aza-2,-deoxycytidine(5-Aza-CdR) on the proliferation and apoptosis of HCC cell line HepG2,and detect the expression of MIA2protein in HepG2after treatment of5-Aza-CdR.Method The proliferation and apoptosis of HepG2treated with0、1、2、4、8、10、 20and40μmol/L of5-Aza-CdR was detected by MTT and flow cytometry test.Then treatedwith0μmol/L and10μmol/L of5-Aza-CdR, the MIA2protein expression was detected byimmunohistochemistry and Western blot.Results Treated with a certain range concentration of5-Aza-CdR, MTT shown that theproliferation of HepG2was inhibited and there was a positively correlation between cellproliferative inhibition rate and drug concentration;Flow cytometry shown that the apoptosisof HepG2was increased and a positively correlation exsisted between early apoptosis rate anddrug concentration;Immunohistochemistry and Western blot displayed that the level of MIA2protein was elevated in10μmol/L group compared with control group(0μmol/L).Conclusion After treated with5-Aza-CdR, MIA2protein expression was enhanced inHepG2, cell proliferation was inhibited and apoptosis was promoted.
