【Objective】To investigate the ratio of HUNK CpG island methylation in tumor tissues and adjacenttissues of the cancer in the patients with colorectal carcinoma and relationship with clinicalpathological parameters.【Methods】The ratio of HUNK CpG island methylation was detected by pyrosequencing in30cases of colorectal carcinoma tumor tissues and adjacent tissues of the cancer; HUNK proteinand mRNA expression was analyzed using immunohistochemistry and real-time fluorescentquantitative PCR (qRT-PCR) respectively. The relationship among HUNK expression andclinicopathological parameters was analyzed.【Results】1.The mean ratio of HUNK CpG island methylation was significantly lower in patientswith colorectal carcinoma(20.67±7.36) than in the healthy control(83.07±2.71)(P<0.05).Low methylation ratio in colorectal cancer tissue was correlated with Patients’ gender, tumorstaging, tumor position and the depth of serosal invasion, but wasn’t correlated with patients’age, the size of tumor and the degree of lymph node metastasis;2.The expression of HUNK was significantly higher in colorectal carcinoma tissues thancancer adjacent tissues as shown by immunohistochemistry (χ2=6.79, P<0.01). HUNKexpression was correlated with Patients’ gender, tumor staging, tumor position and the depthof serosal invasion, but wasn’t correlated with patients’ age,the size of tumor and the degreeof lymph node metastasis;3.qRT-PCR results showed that the HUNK mRAN level of colorectal carcinomatissues was significantly higher than that of cancer adjacent tissues (P=0.020).【Conclusions】1.There was high expression of HUNK in tumor tissues of the patients with colorectalcarcinoma; 2.The expression of HUNK was significantly higher in tumor tissues of colorectal cancerpatients, and it wsa correlated with patients’ gender, tumor grading, the spot of tumor and thedepth of serosal invasion.It might be used as the basis of screening, prognosis determinationof colorectal carcinoma....
