Objective:To investigate the expression and significance of UbcH10in gastric cancer andnormal mucosa tissues.Methods:1. Western blot method protein were used to detect the expressions of UbcH10inthe gastric carcinoma tissues and the cancer adjacent normal tissues2. Reverse transcription polymerase chain reaction (RT-PCR) and Western blotmethod were used to detect the expressions of UbcH10mRNA and protein in43casesof gastric carcinoma tissues and the cancer adjacent normal tissues.3. The expression of UbcH10in117cases of gastric carcinoma tissues,which arecompletely follow-up visited, was detected by using immunohistochemistry4. the patients with gastric carcinoma are follow-up visited by outpatient、hospital、phone、field visit,etc to get information of the patients’ recurrence andmetastasis and death data,and calculate postoperative survival of patientsResults:1. Western blot analysis revealed that the expression of UbcH10protein was alsohigher in gastric carcinoma tissues than in the cancer adjacent normal tissues (P<0.05)2. Real-time PCR results showed that UbcH10mRNA expression of cancertissue increased compared to matching the cancer adjacent normaltissue,respectively(P<0.05)3. The positive rate and adjacent normal mucosa, respectively. The difference ofUbcH10expression was not significant in different groups of the gender of patients,age, tumor site, areas of tumor, while it was significant different groups of tumordifferentiation, invasive depth, lymphatic metastasis and clinical stage (p<0.05). Themoor poorly differentiation and the more increased positive expression. 4. The-5year survival state in gastric cancer with UbcH10high-expressionincreased compared to UbcH10low-expression, respectively.Conclusions:1.UbcH10is upregulated in gastric cancer2. expression of UbcH10considered to be closely related to the prognosis ofpatients with gastric cancer.
