Objective1. Clear Hp infection and the expression of HIF-1α in different gastric mucosa tissueof the patient with gastric adenocarcinoma.2. Explore the relationship between the HP infection and the expression of HIF-1α inpatients with gastric adenocarcinoma of different gastric mucosa tissue and theclinical pathological characteristics.3. Analysis whether there is a correlation between HP infection and HIF-1αexpression in gastric mucosa tissue of the patient with gastric adenocarcinoma.4. Comprehensive analysis the role of HP infection and HIF-1α expression in gastricadenocarcinoma occurrence and development.Methods1. Patients with gastric adenocarcinoma were selected from yanan universityaffiliated hospital in Shaanxi province from January2011to June2012betweenthe ages of25to85for this study.All patients’heart liver kidney and lungfunctions testing have not obvious anomaly before operation,and not for eradicateHP treatment with radiation and chemotherapy. Postoperative were confirmed bypathology of primary gastric gland carcinoma and have a complete clinical casematerial. Using the tissue slice toluidine blue staining method detection selectedresearch object of HP infection status in different organizations of mucousmembrane,application of immunohistochemistry S-P method to test the patients’HIF-1α expression in different tissue of patients with gastric carcinoma.2. Collect the information of the patient with primary gastric gland carcinoma, whichincluded the general characteristics of the population in the study and clinicopathologic feature.3. The data was processed with SSPS17.0software package: using chi-square test tocompare the positive expression rate of HP and HIF-1α in different mucosaltissue;Using spearman rank correlation test to analyse the correlation between HPinfection and the expression of HIF-1α in gastric gland carcinoma tissue.Results1. In this study, there are a total of65patients with gastric adenocarcinoma.Cancerous tissue compared with adjacent gastric mucosa of Hp infection rateswere as follows:56.92%、29.23%. Carcinoma tissues compared with adjacentgastric mucosa, the former has a lower Hp infection positive rate than thelatter(P<0.05), and the difference between the two groups have statisticalsignificance.2. Cancerous tissue compared with adjacent gastric mucosa of HIF-1α proteinpositive expression rate were as follows:61.54%、16.92%. Carcinoma tissuescompared with adjacent gastric mucosa of the HIF-1α protein positive expressionrate, carcinoma tissues has a higher HIF-1α protein positive expression rate thanadjacent gastric mucosa(P<0.01), and there are significant differences in the twogroups.3. The pathological factors relationship to Hp infection including: Lymph nodesmetastasis and TNM tumor staging. Hp infection positive rate of lymph nodemetastasis group is higher than the group without lymph nodemetastasis(P<0.05), and Hp infection positive rate of tumor TNM stage Ⅲ-Ⅳ ishigher than tumor TNM stageⅠ-Ⅱ(P<0.05).4. The pathological factors relationship to HIF-1α protein expression including:Lymph nodes metastasis and TNM tumor staging. The HIF-1α protein positiveexpression rate of Lymph node metastasis group is higher than the group withoutlymph node metastasis(P<0.01), and HIF-1α protein positive expression rate oftumor TNM stageⅢ-Ⅳ is higher than tumor TNM stageⅠ-Ⅱ(P<0.05). 5. There is positive correlation between Hp infection and the expression of HIF-1αprotein in the same cases of gastric adenocarcinoma tissue(P<0.05,r=0.300).Conclusion1. Hp infection and HIF-1α protein expression in gastric adenocarcinoma canceroustissue is an important event to promote tumor growth in the occurrence anddevelopment.2. Hp infection and HIF-1α protein expression are associated with the lymph nodemetastasis and TNM staging of gastric carcinoma, Hp infection positive rate oflymph node metastasis group is higher than the group without lymph nodemetastasis, and Hp infection positive rate of tumor TNM stage Ⅲ-Ⅳ is higherthan tumor TNM stageⅠ-Ⅱ.The HIF-1α protein positive expression rate ofLymph node metastasis group is higher than the group without lymph nodemetastasis, and HIF-1α protein positive expression rate of tumor TNMstageⅢ-Ⅳ is higher than tumor TNM stageⅠ-Ⅱ.3. There is positive correlation between Hp infection and the expression of HIF-1αprotein in gastric adenocarcinoma cancerous tissue,and both may jointlyparticipate in the initiation and progression of gastric adenocarcinoma.
