ObjectiveTo investigate the inhibitory effects of human anti-bFGF antibody onproliferation of human melanoma A375cells in vitro and in vivo, and makepreliminary research on its mechanisms.MethodsHuman melanoma A375cells were cultured. The proliferation inhibition ofhuman melanoma cells was analyzed by CCK-8. The phosphorylation level ofERK1/2triggered by human anti-bFGF antibody was detected by Western Blot. Thexenograft nude mouse model was established to study the inhibitory effects of humananti-bFGF antibody on melanoma in vivo. All nude mice were injected by i.vaccording to the protocol every three days for18days. Body weight and tumorvolume of the mice were measured and analyzed. Microvessel density (MVD) of thetumor tissue was analyzed by immunohistochemistry.ResultsThe results of the proliferation-inhibition experiment in vitro showed that theproliferation of melanoma cells could be inhibited by human anti-bFGF antibody. Theinhibition rate was about24%at the antibody concentration of800ng/ml. The Westernblott results showed that ERK1/2phosphorylation was inhibited by human anti-bFGFantibody, which demonstrated that human anti-bFGF antibody could inhibit theproliferation of tumor cells by inhibiting the ERK signal pathway. The analyse ofbody weight reveals that weight of nude mice in PBS control group, the murineantibody-treatment group, human antibody-treatment group gained, but the nude miceof cyclophosphamide-treatment group lose weight. These results proved that thehuman anti-bFGF antibody is safe. The tumor xenografts nude mice model showedthat human anti-bFGF antibody could inhibit the tumor growth in vivo. The inhibitionrate was about28.12%. The microvessle density of the tumor tissue was also decreasedobviously. ConclusionsThese results demonstrated that human anti-bFGF antibody could inhibit theproliferation of human melanoma cell in vitro and inhibit tumor growth andangiogenesis in vivo.
