ObjectiveA mouse model mimicking transient hypoxia was established to investigate the effectsof transient hypoxia in brain on the cognitive function and the neurogenesis afterlateral ventricular injection of NeuroD in neonatal mouse.Importantly, thisinvestigation may result in novel interventions for management of clinicalhypoxic-ischemic encephalopathy for babies in the perinatal period.Methods1.With reference to Grojean method, n eonatal5d mice were put in100%nitrogenenvironment in a short time(10minutes) to establish the model of transient hypoxia.The neural behavior observation, HE staining were both used to observe brain tissuepathological changes, and evaluate the animal model.2.Experimental neonatal mice were randomly divided into4groups.They areseparately the control group, the hypoxia group, the group of intracerebral ventricleNeuroD peptide injection after hypoxia, the group of intracerebral ventricle NeuroDControl peptide injection after hypoxia. The expression level of NeuroD mRNA andNeuroD protein in mouse brain were respectively detected with RT-PCR andimmunofluorescence staining with BrdU and Doublecortin marked.Results1.During transient hypoxia, the peoformance of neurological behaviors such assystemic cyanosis, creeping around,unconsciousness were found in all of neonatalmice. The result of the traction test, cliff escape experiment and Morris water mazerevealed that the ability of intelligence,spatial learning and memory in the mice modelwere weakened. By means of HE dyeing we found that the hippocampus neuronswere severely damaged after10minutes’ hypoxia.2. The result of RT-PCR showed that NeuroD mRNA expression is visible in braintissue of model mice, but there are no significant differences between groups. 3. The result of immunofluorescence staining appears that there is NeuroD expressionin the brain cortex and hippocampal dentate gyrus, but no significant differencesbetween groups.Conclusions1.By means of putting newborn mice in the environment of100%nitrogen, the modelof transient hypoxia mimicking hypoxic ischemic encephalopathy was set upsuccessfully.2. Through the lateral ventricular injection of NeuroD after transient hypoxia, obviouscognitive improvement is hardly discovered.
