Objective To explore the expression of miR-196and HoxB8in patients ofcolorectal cancer who accepted chemotherapy of Folfox4scheme before theoperation, and illustrating different expression levels of miR-196andHoxB8in patients of colorectal cancer can lead to the discrepantchemotherapy sensitivity with Folfox4scheme or not.Methods Fluorescence quantitative PCR was used to determine theexpression of miR-196and HoxB8in50specimens of neoadjuvantchemotherapy group and30specimens which received the surgery directly.The immunohistochemistry was used to determine the expression of HoxB8in neoadjuvant chemotherapy group, and analyzing the relationship anddiscrepancy chemotherapy sensitivity between miR-196and HoxB8in thesegroups.Results The RT-PCR examination showed that the relative expression levelof miR-196and HoxB8in the group of neoadjuvant chemotherapy were lowerthan the group with direct surgery (P<0.01). Expression of miR-196inthe sensitive group was higher than the insensitive group(P<0.01),however the expression of HoxB8in the sensitive group was lower than theinsensitive group(P<0.05).The Expression of miR-196and HoxB8in thegroup of neoadjuvant chemotherapy has negative relationship, which wasalso exist in the group with direct surgery (the Spearman ratio were-0.595and-0.435respectively).Conclusions The expression level of miR-196and HoxB8were reduced byneoadjuvant chemotherapy with Folfox4scheme. The different expression level of miR-196and HoxB8,which could influence the sensitivity ofneoadjuvant chemotherapy with Folfox4scheme in colorectal cancer. Thehigh level of miR-196can restrain the expression of HoxB8, and thenincrease the sensitivity of chemotherapy with Folfox4scheme incolorectal cancer.
