Objective To investigate the pathological changes of the obstructed kidneys in renalinterstitial injury, and to observe the effects that the marks of epithelial-mesenchymaltransdifferentiation E-cadherin, alpha smooth muscle actin and fibroblast specificprotein-1expression by Ursolic acid intervertion on rat unilateral ureteral obstructionmodels. To provid the experimental basis for clinical prevention on renaltubulointerstitial fibrosis.Methods Twenty-four Sprague-Dawley male rats of180-200g, were randomlydivided into sham operation group (n=6), model group (n=6), model+Ursolic acidintervention group (n=6), model+Enalapril intervention group (n=6). The unilateralureteral obstruction model was established. From the day before operation,intervention group received Ursolic acid and Enalapril ig, sham operation group andmodel group received the same amount of normal saline. The operation renal tissueswere collected14days after operation. HE staining and Masson staining were used toobverse the tubulointerstitial lesions, and Immunohistochemical staining detected theexpression of E-cadherin, α-smooth muscle protein actin and fibroblast specificprotein-1in tubulointerstitial. BCA method detected the total protein content of thesample and correct the ELISA test quantitative detection of alpha-smooth muscleactin expression.Results1. HE staining: Renal tissues were not significant changes in shamoperation group rats. The renal tubulointerstitial lesions of model group rats wereobvious, with infiltrating of inflammatory cells in tissues, swelling of tubularepithelial cells, tubular dilation/atrophy obviously, the tubulointerstitial lesionsdeveloped the fibrosis. Ursolic acid group and Enalapril group the tubulointerstitiallesions were relatively relieved. 2. The semi-quantitative analysis of Masson staining: Compared with shamoperation group, model group and drug intervention group were statisticallysignificant(P<0.05). The difference were not statistically significant among Ursolicacid group, Enalapril group and model group(P>0.05).3. The semi-quantitative analysis of Immunohistochemical staining:①Comparedwith sham operation group, the difference of E-cadherin, alpha-smooth muscle actinprotein and fibroblast specific protein-1expression in model rats group renal tubularepithelial cells were statistically significant(P<0.05).②Compared with model group,the difference of E-cadherin and alpha-smooth muscle actin expression in drugintervention rats group were statistically significant(P<0.05); fibroblast specificprotein-1expression were not significant difference(P>0.05).③Compared withEnalapril group, the difference of E-cadherin expression in Ursolic acid rats werestatistically significant(P<0.05); the difference of alpha-smooth muscle actin andfibroblast specific protein-1expression were not statistically significance (P>0.05).4. ELISA analysis:By BCA detecting the total protein content of the samplecalibrated, compared with sham operation group, model group and drug interventiongroup were statistically significant(P<0.05). The difference were not statisticallysignificant among Ursolic acid group, Enalapril group and model group(P>0.05).Conclution In renal injury process after rat unilateral ureteral obstructionmodels, the marks of tubular epithelial-mesenchymal transdifferentiation E-cadherinexpression was increased, also the expressions of alpha-smooth muscle actin andfibroblast specific protein-1were decreased by Ursolic acid and Enalapril intervention,thereby inhibiting renal tubular epithelial-mesenchymal transdifferentiation, reducingtubulointerstitial fibrosis. Between Ursolic acid and Enalapril have no significantdifference in the inhibition of renal tubular epithelial-mesenchymaltransdifferentiation and reducing the obstructed kidney fibrosis.
