Abstract: Objective: To study the secondary metabolites of Verrucosispora sp.FIM06054from marine sponge and Verrucosispora sp. FIM090395from marinesediments. Methods: Six kinds of fermentation media for Verrucosispora sp.FIM06054and FIM090395were selected to screen the secondary metabolites byHPLC-DAD-MS. The metabolites from the strains were purified by macroporousresin HP-20, silica gel, sephadex LH-20and C18reversed phase columnchromatography. The structure of the compounds was determined by UV spectra,LC-MS, NMR analyses. The cytotoxic activity of the compounds was detected in vitroby MTT assay. Results: Two compounds (06054-1,06054-2) were isolated from theculture filtrate of FIM06054, and one compound (090395-1) from the culture filtrateof FIM090395.06054-1and090395-1were identified as proximicin A and B withsignificant cytotoxicity, respectively. Meanwhile,06054-2was identified as2-hydroxy-3,6-dimethyl-N-(4,5,6-trihydroxy-4-methyl-tetrahydro-pyran-3-yl)benza-mide which was a new compound. This compound was similar to the partial structureof enediyne antibiotic maduropetin. In bioactivity,06054-1showed cytotoxicactivities against tumor cell lines MDA-MB-231(breast cancer cell line), SW-620(colon cancer cell lines), and CNE-2(nasopharyngeal carcinoma cell line) with IC50values of80.17,46.00and55.56μM, respectively.06054-2showed cytotoxic activitiesagainst CNE-2cells with IC50values of22.12μM, weak activities againstMDA-MB-231, SW-620and SMMC-7721(hepatocarcinoma cell line) cells.090395-1showed cytotoxic activites against K-562(leukemia cell line) cells with IC50valuesof62.20μM, weak activity against MDA-MB-231and CNE-2cells. Conclusion:Theabove results indicated the genus of Verrucosispora had potential to produce novelsecondary metabolites with significant bioactivity.
