OBJECTIVE The aim is to detect the relationship between parameters of DCE-MRI and theexpression of HIF-1α and CA IX in fresh tissues of nasopharyngeal carcinoma(NPC),and toexplore the correlation of parameters of DCE-MRI,the expression of HIF-1α and CA IX in NPCtissue and patients’ clinical biological behaviors.METHODS62patients of NPC were subjected to DCE-MRI scanning and DCE-MRIparameters were obtained. Do nasal endoscope in marked areas of the patients.And theexpression of HIF-1a and CA IX were measured by means of immunohistochemistry. The mutualcorrelation between DCE-MRI parameters and HIF-1a、CA IX and the role in NPC was alsoanalyzed.RESULTS1. Among the DCE-MRI parameters of the62NPC patients, no significantcorrelation were obtained between T1on set and patients’gender and ages. But T1on set weresignificantly correlated with T-stage,N-stage and clinical stage respectively(P<0.05)The higherthe T-stage,N-stage and clinical stage was,the longer the T1on set was,and the differencesamong the different were statistically significant(P<0.05).No significant correlation wereobtained between MS and the patients ‘s gender and ages,but MS were significantly correlatedwith T-stage,N-stage and clinical stage respectively(P<0.05). The higher T-stage,N-stage andclinical stages was,the higher the MS was. And the differences among the different stages werestatistically significant(P<0.05). No significant correlation were obtained between TTP, AUC andclinical-pathological parameters(P>0.05).2. The positive expression rate of HIF-1α was79.0%in NPC,0%in chronic nasopharyngitis respectively, and there was significant difference in theexpression of HIF-1α protein between NPC and chronic nasopharyngitis tissue(P<0.05). Theexpression of HIF-1α protein were correlated significantly with patients’ age(≥50y vs.<50y),clinical stage and lymph node metastasis (positive vs. negative)(P<0.05).The expression of CAIX protein were correlated significantly with patients’ age and N-stage(P<0.05), but nosignificant correlation were not observed between HIF-1α expression and patient’s gender,T-stage and clinical stage(P>0.05).3、The positive expression rate of CA IX was83.9%in NPC,0%in chronic nasopharyngitis respectively, and there was significant difference in theexpression of CA IX protein between NPC and chronic nasopharyngitis tissue(P<0.05). The expression of CA IX protein were correlated significantly with patients’ gender andN-stage(P<0.05), but no significant correlation were not observed between CA IX expressionand patient’s age, T-stage and clinical stage(P>0.05).4. The positive expression of HIF-1αprotein was significantly correlated with T1on set(P=0.015) and AUC(P=0.001),but nosignificant correlation were observed between the expression of HIF-1α and MS、TTP(P>0.05).The positive expression of CA IX protein was significantly correlated with AUC(P=0.000),butno significant correlation were observed between the expression of CA IX and T1on set、MS、TTP(P>0.05).CONCLUSIONS1. Among the DCE-MRI parameters,T1on set, MS were significantlycorrelated with T-stage, N-stage and clinical stage in NPC patients(P<0.05).The higher the T1onset and MS were, the higher the T-stage,Nstage and clinical stage were,which support that T1onset, MS can be an important measurement for diagnosis,staging and prognosis of NPC.2. Thepositive expression of HIF-1α and CA IX in NPC is significantly different from that in chronicnasopharyngitis,which support that HIF-1a and CA IX played an important role in the progressand metastasis in NPC, and can be an important measurement for diagnosis, staging andprognosis of NPC.3.Parameters of DCE-MRI(T1on set、AUC)were significantly correlated withthe positive expression of HIF-1a. AUC were significantly correlated with the positiveexpression of CA IX, which support the parameters of DCE-MRI can be an importantmeasurement for hypoxia status, diagnosis, staging and prognosis of NPC.
