Objective: To investigate the therapeutic effects and acting mechanism ofChangyanqing in rats with ulcerative colitis (UC) induced by immune-combinedTNBS/ethanol.Methods:60healthy rats were divided into2groups. Normal control group wasinjected saline and model group was established with immune-combined TNBS/ethanol.Four rats were sacrificed to observe the macroscopical focus of infection in bowels in21day and UC model was taken out. The UC model rats were randomly divided intomodel control group, Changyanqing group (40g·kg-1), Changyanqing group(10g·kg-1)and SASP group(0.1g·kg-1). All model rats were treated for4weeks. During the4-weektreatment, disease activity index (DAI) were observed. At the end of treatment,Morphological observation was carried out on the rats’ colon and lung (colonic mucosaldamage index (CMDI) scores and histological score (HS)); The activity ofmyeloperoxidase (MPO) was measured by biochemical method; IL-6, IL-8, IL-17, sIgAand TNF-α was determined by ELISA; P-selection was determined byimmunohistochemical staining (SP method).Results:(1) DAI, CMDI, HS, MPO in model rats were obviously increased(P<0.01);IL-6, IL-8, IL-17and TNF-α were markedly higher in blood serum(P<0.01);P-selection was also markedly higher in colon(P<0.01); whereas sIgA(lung and colon)was significantly decreased in UC rats compared to normal rats(P<0.01).(2) Comparedwith the UC model group, Changyanqing(10g·kg-1and40g·kg-1)DAI, CMDI, HS(lungand colon), MPO were obviously reduced (P<0.01or P<0.05);(3) IL-8, IL-17andTNF-α was markedly reduced in Changyanqing(10g·kg-1and40g·kg-1) group comparedto UC model(P<0.01); Changyanqing (40g·kg-1) group was also obviously reducedIL-6compared to UC model(P<0.01);(4) Compared with the UC model group,Changyanqing(10g·kg-1and40g·kg-1) sIgA(lung and colon) were significantly rised in the rats(P<0.01);(5) P-selection in the colon was markedly reduced inChangyanqing(10g·kg-1and40g·kg-1) group compared to UC model(P<0.01).Conclusion:(1) Changyanqing can relieve symptoms of UC model rats;(2)Changyanqing improve the immunity ability by increasing the sIgA in the lung andcolon;(3) IL-6, IL-8, IL-17and TNF-α were reduced in blood serum to alleviateinflammatory response at Changyanqing group;(4) P-selection was decreased in colonby Changyanqing group.
