Due to the rapid growth and aging of the population, the increased urbanizationand the changes in the life style, the prevalence of the obesity and its complications,such as hyperlipidemia, stands global increase in recent years. Previous studies havesuggested current available treatments of obesity without drugs to be palliative andunsustainable. Thus, the developments of the valid and safe drugs against the obesityshow great urgency.Psidium guajave, which belongs to the myrtle family and guava genera, distributeswidely in tropics and subtropics and is also largely planted in Guangdong, Guangxi,Fujian and Taiwan in China. Fundamental evidences have demonstrated the multiplepharmacodynamic activities in the Psidium guajave leaves which may providetherapeutic potential to the cure of obesity and its complications. The therapeuticpotential was further confirmed by our previous studies that quercetin-3-O-(6”–feruloyl)-β-D-galactopyranoside extracted from Psidium guajiave leaves coulddramatically suppress the adipogenesis of the3T3-L1cells preadipocytes.In our study, we mainly investigate the anti-obesity and lipid lowering effects ofthe extracts from the Psidium guajave leaves in high-fat diet induced obese mice. Theresults suggest that extracts from the Psidium guajave leaves can substantially lead tothe weight loss and inhibit the increase of lipid. What’s more, with a certaindose-response effect, these effects became more obvious when using a higher dose ofthe extracts, making the200mg/Kg the most effectual dosage. Parallel experimentsusing fenofibrate serve as positive control. As it was previously reported that PPARγ2and C/EBPα played important roles in the regulations of adipogenesis, we furtherstudied how these two factors involved in the suppressive effects on obesity anddyslipidemia by the extracts from Psidium guajave leaves through performing real-timePCR assays. Our study demonstrated that the expression of these two factors waslargely suppressed and the maximal suppressive effects appeared at the dosage of200mk/Kg. Briefly, we proposed a novel, promise candidate drug for the cure of obesityand its complications.
