Objective:1. To observe the expression changes of EGFR and KRas proteins in epihtelialovarian carcinoma and their clinical significance.2. To analyze the incidence and profiles of EGFR extracellular domain IVmutations in epithelial ovarian carcinoma,which provide gene detection methods andtheoretical basis for the targeted treatment.3. To investigate the relationship between the expression of EGFR and themutations of EGFR extracellular domain IV.Methods:1. The expression of EGFR and KRas proteins in45cases of epithelial ovariancancer,18cases of benign ovarian cystoadenoma and30cases of normal ovarian tissuewere detected by SP immunohistochemical method.2. Randomly selected45cases of Paraffin-embedded tumor tissues and15casesof fresh ovarian cancer specimens,using routine phenol-chloroform method forgenomic DNA extraction,appliying polymerase chain reaction and direct sequence forEGFR extracellular domain IV14,15exon mutation.Results:1. The expression of EGFR and KRas proteins in ovarian cancer was marklyhigher than that in benign and normal ovary tissues(P<0.05). The expression rate ofEGFR was in correlation with clinical stage(P<0.05).The expression rate of KRas wassignificantly in connection with clinical stage,historial classification(P<0.05). Therewas positive correlation between EGFR and KRas,correlationcoefficient is0.613(p<0.05).2. No EGFR extracellular domain IV14,15exon mutations were detected inepithelial ovarian carcinoma by PCR and direct sequence.Conclusion:1. The expression of EGFR and KRas is obviously up-regulated in epithelial ovarian cancer tissues,which have positive correlation. Both genes may play animportant role in the generation and development of ovarian carcinomas.2.The positive expression rate of KRas is higer in the early or highdifferentiation ovarian cancer tissue. So KRas may act as a promoter gene of ovariancancer.3. There was rare or no EGFR mutation in exon14and15in epithelial ovariancarcinoma.There may be other EGFR potential or unknown mutation points.4. The abnormal expression of EGFR protein has no relation to EGFR mutationwhich maybe not the main mechanism of inducing overexpression.
