Objective to investigate AMPA receptors expression in olfactory bulb(OB) ofchronic rhinosinusitis with dysosmia, to discuss the pathogenesis of chronic rhinosi-nusitis with dysosmia.Methods: SD rats were inoculated intranasally streptococcus pneumoniae toestablish chronic rhinosinusitis with dysosmia model. Histological changes ofolfactory mucosa in the model were examined by hematoxylin and eosin stain.Determination of olfactory function Was carried out via the procedures of buried foodpellet test(BFPT)among these modeling rats, The GluR2of AMPA receptors inolfactory bulb were determined by reverse transcription polymerase chain reactionmethod (RT-PCR)and Immunofluorescence technique.Results: Various degrees of inflammatory pathology could be determined by thetissue samples of olfactory mucosa among all the rats in control group and modelgroups,presenting a changing process from acute to chronic,including thickerolfactory mucosa, increased glands,squamous-like metaplasia, in addition the numberof neutrophils, plasma cells, lymphocytes increased, lymphocytes were predominantin later chronic procedure.The datas which shows the damage of olfactory functiondetected by BFPT among the model rats was significantly statistical implications(P <0.01)when compared to control group. Only the12group shows dysosmia.RT-PCRshows AMPA receptor GluR2mRNA expression in12weeks group declinedobviously when compared to the other3groups (P<0.05). GluR2immunoreactivityshowed heavy staining in the bulbs of12model group rats comparing with3othergroups,which was similar with results of RT-PCR.Conclusion:The chronic rhinosinusitis with dysosmia model can be successfullydeveloped by the procedures inoculationg streptococcus pneumonia into nasal cavityof rats.the expression of AMPA receptor is downregulated in OB in the chronicrhinosinusitis with dysosmia rats,AMPA receptor may play a crucial role in the pathogenesy of sysosmia which induced by CRS.
