Edema disease of swine, also named intestinal toxemia, is extremely harmful to the pigindustry. Previous studies demonstrated that the major virulence factor of Swine Edema disease isShiga-like toxin IIe(SLT-IIe). Skullcap Injection is made up of Radix Scutellariae after extraction.Its main composition is Baicalin. Radix Scutellariae is one of the commonly used herbs in China,which can clear fever, release fire, neutralize poison and stop bleeding. It has extensivepharmacological actions and important clinical values. Baicalin is flavonoids isolated from theRadix Scutellariae’s root with the effect of antibacterial, anti-inflammatory, improvemicrocirculation. So our study aimed to investigate the effects of Skullcap Injection on porcineedema disease, and attempted to evaluate the safty and effectiveness according the veterinary drugstandard.Firstly, we assessed the effects of Skullcap Injection on SLT-IIe-induced Vero cellcytotoxicity. In vivo study, SLT-IIe was given to mice by intraperitoneal injection to estiblishedema disease model. The therapeutic effect of Skullcap Injection was reflected by the index ofsurvival rate, pathological changes, histopathological changes. We found that Skullcap Injectionsignificantly inhibited SLT-IIe-induced Vero cell cytotoxicity in a dose manner, reduced themotality rate of edema disease in mice model by SLT-IIe-induced and significant improved thehistopathological changes of intestine and liver.The safety of Skullcap Injection was evaluated by acute toxicity test and muscular irritationtest. To observe the acute toxicity reaction and death of mice one day after intraperitoneal injectionof Skullcap Injection so as to appraise its safety. Skullcap Injection was given to mice by3timeswithin24h, the maximum dosage of Skullcap Injection was1.8ml/20g (converted baicalin was4.5g/kg body weight), the Skullcap Injection is relatively safe. In the stimulating test, the rabbitwere given Skullcap Injection by injectioned on quadriceps for1mL. The result showed thatSkullcap Injection is safe which be available for intramuscular use.Finally, porcine edema disease model was established by means of intraperitoneal injection ofE. coli O139in piglets. Through clinical symptoms, pathological autopsy, pathogen separation andidentification, regression test and the cure rate index to judge the model is set up successfully andthe therapeutic effect of the injection. The result show that the porcine edema disease model isestablished successfully and the therapeutic effect of Skullcap injection is good. Experimentalclinical trials identified the swine edema disease clinical recommended dose was intramuscularinjection, per kilogram of body weight with0.2mL,1time a day for5days. In summary, Skullcap Injection can protected Vero and has a good therapeutic effect onSLT-IIe-induced porcine edema disease and has higher safety. Identified the swine edema diseaseclinical recommended dose was intramuscular injection, per kilogram of body weight with0.2mL,1time a day for5days. The conclusion lay the foundation of the further clinical trials and clinicalapplications.
