Porcine epidemic diarrhea virus (PEDV) is an acute and highly contagious entericdisease characterized by severe diarrhea, vomiting, dehydration, and a mortality rate of up to90%in suckling piglets. Recently, the outbreak of PEDV infection shows an increasing trend inworldwide. basic research regarding the pathogenesis of PEDV has important significance.In this study, the differential expressed protein of the PEDV-infected Vero E6cells wasinvestigated using an isobaric tag for relative and absolute quantitation (iTRAQ)-MS/MSapproach. The results showed that the numbers of identified peptides and proteins betweenPEDV-infected group (V) and PEDV-infected mock (C) were15564and3178, respectively, ofwhich3171proteins have quantitative information. A total of126differentially expressedproteins (DEPs) were found between the V group and C group (P<0.05, ratio≥1.2), andmolecular function of the DEPs mainly focused on the binding and catalytic activity accordingto Gene Ontology categories. The pathways, which the DEPs participated in, involved inmetabolism, organismal systems, cellular processes, genetic information processing,environmental information processing and diseases, of which some anti-viral pathways andproteins, such as RIG-I-like receptor signaling pathway, Rap1signaling pathway, autophagysignal pathway, MAPK signaling pathway, PI3K-Akt signaling pathway, Jak-STAT signalingpathway, and integrin β2/β3, cystatin-3, were highlighted as potential factors in PEDV infection.Western blot results showed that the ratios of the up-regulated protein integrin β3between theV and C groups were consistent with those obtained using the quantitative proteomics analysis.The resulting data will provide valuable information to increase understanding of PEDVinfection.
