As different group of microorganism can produce different PLFA which is the important constituent of the living cell membrane phospholipids by the corresponding biochemical pathway, it makes part of PLFA always appear in the same group of microorganism. So the PLFA which is regarded as the basis of dividing the Living microbial community becomes the main base. The text studies the PLFA technic and the Analysis of expansion and the spectral data of Sherlock MIS system function as the main line to probe into the way of PLFA extraction, analysis and data processing models. It also compares the functional relation of Microbial Community Structure between soil and litter fall. The results are as flow:(1) The way of PLFA extraction has been constructed and improved which is so efficient by contrast with Oscillation extraction, and it solves the problem of pre-processing of the multitudinous samples.(2) Selecting ultrasonic time, ultrasonic temperature and the Extraction of dose as the main factor of PLFA extraction. Regarding the extraction percentage as response value and get the extraction technic improved by connecting the response value.(3) By collecting the forest soil and litter fall in Mao Xian, regarding the PLFA as the Indicator of specific microorganisms on the dividing of biocoenosis and using the interrelationships by analyzing the microorganism groups in soil and litter fall, the results indicates that the whole diversity in soil and litter fall is not notable. And the result of the Aerobic bacteria and gram negative bacteria is prefer soil while other group of microorganism is prefer litter fall can also be reached. At the same time, it shows that ultrasound has little influence to the extraction of pathogenic bacteria and aerobic formers, but has no influence on the whole microbial Community Structure, by using the major constituent to analysis the microbial Community Structure.(4) Taking a certain range of concentration C19as the interior label and sample size of60ã€100ã€120ã€200ã€240ul to take the test, it has proved that internal standard sample size has directly influenced the result of PLFA microorganism. And by the assessment of unknown samples, the internal sample Range has been ascertained which reduces the deviation by the internal standard sample size.(5)The two common methods of PLFA data processing are compared to reach the best one in this text, and correlating with instrument condition to put forward a reference processing method, and correlating with instrument condition to improve the processing method. at last relying PLFA as the Indicator of specific microorganisms writing spectrogram data-analytics programs to improve the efficiency and accuracy of data processing, and achieving the data processing mode of ecological analysis. |
