| AM symbioses and root nodule symbiosisare two special symbiotic form. With the study of the symbiosis mechanism, people get some consensus on the evolutionary relationships between them. Root nodule symbiosis is to be evolved from AM symbiosis, Genetic loci of the plant genome between root nodule symbiosis and AM symbiosis signaling pathway signaling pathway exist overlap. For example, part of the rice genes regulate AM symbiosis functioned in root nodules symbiosis of legumes, there are at least five rice genes that regulate AM symbiosis play a role on both kind of symbiosis. This became an important basis for exploration that transfer the root Nodule symbiosis system to non-root nodule symbiosis plant. In recent years, the researchers had cloned a series of key symbiosis genes by using the different mutants of symbiotic defects of model legume(Lotus japoncius and Medicago truncatula). Based on the resules, people established the model of nodulation signaling transduction pathway. we try to transfer these key symbiotic genes into rice o establish a nodulation signaling transduction. Transformed the pCAMBIA1302, pCAMBIA1302-35sNSP1-35sNSP2, pCAMBIA1302-35sNSP1-35sNSP2-35sNIN and pCAMBIA1302-35sNSP1-35sNSP2-35sNIN-35sLHKl into Nipponbare through callus, and get a lot of transgenic pedigree.3. Ten days after inoculation, both transgenetic group and control can observe rhizobium invasion of the epidermis and root hairs and form structures similar to infection threads. Almost all of which have appeared beside the lateral root.4. Rhizobium inoculation changed the apical portion of the lateral root development of rice. The number of lateral root primordias and lateral root incressed significantly after rhizobium inoculation than the uninoculated rice respectively both transgenic rice and control. After inoculated with rhizobium, the lateral roots number of transgenic group was significantly greater than the control group.5. The expression of some symbiotic gene orthologs of transgenic group have no big difference transgenic group after inoculated with Rhizobium. And OsRR5, which is a hormone-induced gene, is up-regulated. |
