Abstract/Summary:
Tissue morphological method is used to observate the uterus, fetus and placenta in sows of different periods of pregnancy in this study.The immunohistochemistry is used to detect and analyze the expression of PPAR-y in the placenta. The role of PPAR-y agonist in proliferation of swine umbilical vein endothelial cells is detected by cell immunofluorescence and cell proliferation assay technology.The following results were obtained.1. The eye view change of the development in sows’uterus and tetals during the pregnancy.The significant growth of uterus of25days and40days pregnancy has been observed. The length of pregnancy uterine horn changes in the range of90to173cm due to the number of the fetus. The amniotic sac and urine amniotic sac has developed and the fetal body has been basically formed. The variation degree of the fetal weight at25days of gestation is13.1%; The variation degree of the the fetal weight and body length at70days of pregnancy is respectively24.5%and9.5%. The fetal inter-individual difference in body weight is greater than the degree of difference of body length.2. The histological observations of uterus and placenta in pregnant sowsThe curving part of the uterine mucosa begins to form a structure like fluff halo at25days of gestation; At40days of gestation, uterine mucosal gets hyperplasia.Compared to25days of pregnancy, the uterine mucosa is is more developed and dense. Besides, the blood vessels that close to the mucosal epithelium have increased. The bands formed by secretions in the gap between the uterine mucosal epithelial cells and trophoblast cells can be observed at high magnification.3. PPAR-γ gene in pregnancy25days and40days sow placenta expression of pregnancyPlacental tissue PPAR-y immunohistochemistry experimental results show that at25days of gestation, PPAR-y showed strong positive marker in luminal epithelial cells in the maternal blood vessels, weakly positive mark, almost no mark on the gland;40days of pregnancy in the gland, PPAR-y endothelial cells and luminal epithelial cells on presents strong positive mark, maternal blood vessels rendered or strong or weak positive mark. This shows that, PPAR-y in the glandular cells do not express about pregnancy25days and they only began to express PPAR-γ at about pregnancy40days. Within forty days of pregnancy, PPAR-γ were expressed in the luminal epithelial cells and maternal vascular.4. The swine umbilical cord vein endothelial cells cultured in vitro experimentsThe primary pig vascular endothelial cells are bright spherical and exist in groups after being detached; Once they turn to the adherent cells, their shape become spindle or polygonal with the central nucleus protruding and less individual cells and most of the cells exist like islands.When the cells grow to a monolayer, they will show the typical "cobblestone"-like arrangement.5. The role of PPAR-y agonist in proliferation of swine umbilical vein endothelial cellsThe umbilical vein endothelial cells are collected from the gestating sows. After being cultured in vitro, purified, passaged and identification, the cells are respectively exposured to the2.5μmol/L,5μmol/L,10μmol/L,20μmol/L,40μmol/L and80μmol/L of rosiglitazone.Every6hours,12hours and24hours the cells were measured the absorptiometry values at450nm with a microplate reader. It is found that the concentration of5μmol/L rosiglitazone has a more significant role to promote the growth and the reproduction of the swine umbilical vein endothelial cells.
