In this study the role of chlorogenic acid in lipid metabolism with long-term high-sucrose-high-fat diet in Sprague-Dawley (SD) rats, and by the detection of physiological and biochemical and gene expression, and to explore its mechanism.In this study, after one week acclimatizing feeding forty male SD rats (130.36g±2.68) were randomly divided into four groups,10rats/group:the normal control group (NC), the high-sucrose-high-fat diet control group (HSF), the high-sucrose-high-fat diet with low-dose chlorogenic acid group (HSF-LC) and the high-sucrose-high-fat diet with high-dose chlorogenic acid group (HSF-HC). All rats of the control group were fed a normal chow diet and the others were fed high sugar high-fat diet, while HSF-LC group and HSF-HC group were given20mg/kg and90mg/kg chlorogenic acid by gastric perfusion respectively, the others were given placebo by gastric perfusion once a day. Water and foods were available to the animals ad libitum. The body weight of rats were measured weekly, food consumption was monitored daily. The serum content of total cholesterol (TC), high density lipoprotein cholesterol (HDL-c), low density lipoprotein cholesterol (LDL-c) and triglycerides (TG) were determined using automatic Biochemical Analyzer. Expression of PPARa, PPARy either in liver, intestine or adipose tissue, and expression of Acetyl-CoA carboxylase2(ACACB) in liver were analyzed using quantitative real-time PCR. Livers tissue sections were stained with H&E method for normal histological assessment.Results showed rats visceral fat weights and contens of serum lipid were higher in HSF group than NC group. Both chlorogenic acid interference groups with low dose and high does had lower visceral fat weights and contens of serum lipid than HSF group. In liver, compared with the NC group, the expression of PPARa mRNA were down-regulated and the expression of PPARy and ACACB mRNA were up-regulated in HSF groups; compared the HSF group, the expression of PPARa mRNA were up-regulated and the expression of PPARy and ACACB mRNA were down-regulated in the two chlorogenic acid interference groups. However, in adipose tissue, PPARy expression level had an opposite tendency among the in the two chlorogenic acid interference groups. In intestine, compared the NC group, the expression of PPARa mRNA were decresed and the expression of PPARy mRNA were incresed in HSF groups; compared the HSF group, the expression of PPARa mRNA were incresed in the two chlorogenic acid interference groups, but PPARa mRNA were incresed in HSF-LC group and decresed in HSF-HC group. Images of tissue sections showed that in the HSF group rats, the liver cells were filled with more fat droplets and macrophages accumulated in adipose tissue; however HSF-HC group significantly ameliorates these symptoms.All together, our data suggested that CGA might inhibit lipid synthesis in liver cells intestine and adipose tissue by regulating genes, rescue rats from developing obesity and fatty liver induced by high fat diet.
