In this study, seven breeds/combination of two exotic breeds Duroc, Large White, four local breeds Shaziling pig, Wuzhishan pig, Daweizi pigs, Taoyuan black pig, as well as DLY(Duroc×Yorkshir×Landrace) of401pigs in total as experimental materials to study insulin-induced gene2(INSIG2) and free fatty acid receptor1gene (GPR40) single nucleotide polymorphisms,the genetic polymorphism of INSIG2genes and GPR40gene were analyzed by PCR-RFLP method, and four variable sites and pig meat quality traits associated. The main results are as follows:Use directly sequenced of the PCR products combined with PCR-RFLP method detected INSIG2gene-502locus Tâ†'C mutation,-460locus Gâ†'C mutations and-609locus Tâ†'C mutation. And Compared the GPR40gene PCR product sequencing results, it is found that the GPR40gene exon region606locus exist Aâ†'G mutation.Detected the gene INSIG2-502Tâ†'C, INSIG2-460Gâ†'C, INSIG2-609Tâ†'C three locus in the distribution of seven different breeds, found that Shaziling pig do not exist polymorphism in the three seats, the Wuzhishan pig and Daweizi pig have no polymorphism in locus INSIG2-460Gâ†'C, other breeds in these three loci all have polymorphism. GPR40gene606Aâ†'G test results found that Shaziling pig has no polymorphism, and the remaining six breeds all have polymorphism.The results shows that INSIG2-502Tâ†'C loci, TT genetype significantly higher than the CC genetype in Water holding capacity and Tenderness; INSIG2-460Gâ†'C loci, the CC genetype and GG genetype hace no significant difference in storage losses, while the CG genotype was significantly higher than the CC genetype and GG genetype, the marbling GG genotype and CG genotype is not significant but were significantly higher than the CC genotype, fleshy chroma GG genotype and CC genotype difference was not significant, but significantly higher than the CG genotype, CC genotype and CG genetype have no significant difference; INSIG2-609Tâ†'C loci TT genotype storage loss is significantly higher than that of CT genotype, but no significant difference and CC genotype, tenderness traits CC genotype was significantly higher than the CT genotype and TT genotype.The results showed that the GPR40gene606Aâ†'G seat storage losses AA and GG genotype difference was not significant, but the AG-significantly lower than AA type with GG type; flesh-colored levels and meat quality chroma same, AA and AG significant nature of higher than the GG genotype AA and AG were no significant differences.The demerit of this study show that the INSIG2gene maybe be used as a molecular genetic markers of pig water holding capacity, tenderness and marbling; GPR40gene maybe be used as a molecular genetic markers of a flesh-colored.
