| The Bacillus subtilis ACCC-10619was purchased from preservation center of Academy of Agricultural Sciences of China,this subtilis has high productivity of spore and super degradation for pig manure compost. Which makes it has bright future for application. The preliminary study on the effects of composting of manure testing and evaluation,after fermentation and condition optimized, the main result are as follows:(1) The optimalculture media and fermentation conditions for Bacillus subtilis ACCC-10619were determined by methods of single factor tests and orthogonal experiments. The results showed that the optimal conditions were as follows:wheat bran3.0%, glucose3.0%, ammonium sulfate0.5%, manganese sulfate0.05%, initial pH7.0, cultured at33℃for36h with static time12h. The output amounts of spore reached8.5×1010cfu/mL under volume of10L expanding fermentation. It would provide reference for production applications of Bacillus subtilis microbial ecological agents.(2) The fermentation and conditions by shaking culture were optimized for enlargement production. The quantity of spore reach to8.50x1010/ml. The quantity of Spore in liquid fermentation were increased, the biological control techinique were improved and production cost was reduced. The study provide technology data for commercial production and large scale application of Bacillus subtilis in Agriculture.(3) Bacillus subtilis ACCC-10619were applied to degrade pig manure composting. By adding10%straw powder on fermentation heap, with2%inoculum (ratio by weight), then inoculated Bacillus subtilis ACCC-10619accordingly, Set Initial watery of composting at60%and stir the heap once every three days. Fermentation period of30days,the results show that,adding staw and bacillus subtilis ACCC-10619during pig manure composting,degradate speed of fermentations was faster and the effect was batter.The maximum temperature reached67.3℃,the highest PH value reached8.98, Water content reduced by34.2%, C/N decreased by13.2.which indicated that the composting reached maturity level. |
