Study On Phytic Acid And Phytase In Barley

This research studied the phytic acid determination method in barley,compar kinds of phytic acid content in domestic and overseas. Results show:phytic acid content in domestic is higher than overseas.It also studied phytic acid storage part,change of phytic acid content in germination.At the same time,it confirmed the relationship between phytic acid content and protein content. Finally show:phytic acid is mainly kept in barley endosperm,the content is reducing with germination and it is bound up with protein content,especially prolamin.Phytate is a composite which is hard to be soluble.So it has seriously affected the solution and extraction ratio.Because the phytase content and vitality is low in barley,we considered adding exogenous phytase. One hand,use the phytase to resolve phytate to increase malt solution. On the other hand,owing to the phytase is the first enzyme to be activated and it has influence on other hydrolase,so we studied adding phytase in malting had made what affection on amylase、protease、phytase and β-glucanase.At the same time,we payed attention to the change of malt extraction ratio、pH、β-glucan、kolbach index and α-amino nitrogen etc.Results show:adding exogenous phytase increase the activity of amylase、protease、 phytase and β-glucanase.And in the process of decomposing,phytate release phosphorous ion.It satisfies the energy need in germination,makes barley grow exuberant,cuts short the germination cycle,makes malt extraction ratio increase1%.Then use this method in the factory to make enlargement experiment.Results show:it makes malt extraction ratio increase1%,improving pH、β-gluca、kolbach index etc malt quality index.Frome theoretical side,this research discovered the reason for affection to hydrolase activity.Because the gibberellin can induce enzymatic synthesis, it plays a part in barley by the phytase.By using HPLC determine two kinds of barley gibberellin content.Results show: gibberellin content in barley which had been added phytase is higher.So it comes to a conclution:adding phytase stimulate gibberellin content to increase.Gibberellin induce enzymatic synthesis and activate zymogen.So it increases enzyme activity,speeds up germination,cuts short the malting cycle,increases malt extraction ratio,improves some key malt quality index.
