Petunia hybrida is one of the model plant of floral organ development research and isalso important ornamental flowers.Petunia hybrida due to relatively easy regeneration,simpletissue and cell manipμLation techniques, short life cycle, clear genetic background, becomethe model plant of floral organ development research.Abstract MADS box gene pMADS4was isolated from Petunia hybrida by RT-PCR. TheresμLt of the sequencing showed that this nucleotide sequence of pMADS4gene has100%identity with the published sequences. The gene is910bp in length, with an open readingframe encoding a253amino acid protein, which displayed the structure of typical plantMADS box protein and is close similar to ZAG3of Zea mays, DlMADS18of Dendrocalamuslatiflorus and AGL6of Arabidopsis thaliana by64%,64%and61%, respectively. TheseresμLts suggest that pMADS4gene be a AGL6homolog and might have the same function asother AGL6homologous genes, which might promote flower and regμLate floral organformation.Then through the genetic transformation, RT-PCR detection of transgenic plants, analysisof conversion plant phenotype and the condition of in vitro morphogenesis to determine thePetunia hybrida AGL6homologous gene function.such a transformation also confirmed byRT-PCR detection of FBP1expression, a Petunia gene specific expression to corolla andanthers.1.Under the long-day conditions the transgenic plants flower earlier than their wild types.By the phenotypes, the transgenic Petunia plants show earlier flowering cμLtured in soilmedium, and extreme earlier flowering phenotypes which the plants with3petal-like leavesappear a ovary-like structure at terminal in vitro.These instructions Petunias AGL6homologous gene has flower meristem characteristics, and participate in plant flowering time.2.Single pMADS4transgenic plants converted their leaves to petal-like in vitro, and theleaves of weak phenotypes produce abnormal calli, and further bud differentiation moredifficμLt, Petal-like structures, rod-like structures (probable anthers), ovary-like structuresand buds can produced directly from the transgenic calli on bud differentiation medium(BDM). These ovary-like structures also can be produced from the shoot terminal of transgenic plants and calli on rooting medium (RM). But in all the process, homologuetransforming does not appear. The above phenomenon, Petunias AGL6homologous bringabout homoeosis leaves and flower organs, Illustrate Petunias AGL6has the function offlower organs nature.3.Genetically modified petunia plant flower organ development is not normal, lead toovule abortive and cause plant strong ability serious decline. Shown above, Petunias AGL6homologous genes involved in the four kinds of flower organs and ovule development, has theeffect of flower organs and ovule development function.
