Preadipocyte is one of the main cell of connective tissue, it is also the main cellof adipose tissue. It is from embryonic mesoderm and has differentiation ability. I tparticipates in the formation of mature adipose tissue, and can differentiate to adipocyte in some situations. Because of preadipocyte has differentiation ability, it becomes the source of pluripotent stem cell. It is the main cell model of adipose tissue study.The shape of preadipocyte is oval to round. Nuclei is in the middle or at the side, the cell body increases gradually. The cell is fusiform, cytoplasm has small lipid droplets. But the literature of the morphology of macrophages describes it as round, oval and a polygonal or irregular shape. The surface of macrophage has slightly villous bumps.The nuclei is often in one side with sag and is oval. Macrophage is the main cell of connective tissue. Foreign research has shown that preadipocyte can turn to macrophage in some situations and they have a striking similarity in profile. Therefore, this study aims to explore the ultrastructural morphology of preadipocytes in the mousecervix and rule out the possibility of macrophages.This experiment used kunming mice and macrophages as the research object, tookdelivery cervical tissue in mice, made frozen section and did oil red O stain test, ascertained the small lipid droplets distribution in mice cervix, so to locate transmissionelectron microscopy semi thin section and find cells which contain a small fat dropsquickly. And their morphological features are hyperchromatic nuclei, a large numberof small fat drops and abundant mitochondria. Cells also have microvilli protrusion. Doing immunofluorescence experiments to macrophages and locate them, therefore makesure the more accurate location in cervical connective tissue.The first working fluid resistance is F4/80,the second working fluid resistance isFITC. The experimental resultis that macrophages are under the cervical mucous membrane layer.Through the observation to the frozen section oil red O staining and ultrastructural which from delivery the cervical tissue cells in mice and macrophages immunofluorescence experiments, we can conclude that preadipocyte and macrophage are not inthe same location in the cervical. That is the cell under cervical mucosa basement co nnective tissue in mice is preadipocyte instead of macrophage. By comparing macrophage with preadipocyte microstructure, we can find that they both have large nucleus and pseudopodia, but there is no lysosome in preadipocyte, there are all levels of lysosome in macrophages.
