Heat stress is a major factor affecting the production of chicken, this study isbased on pre-study analysis of microarray expression profiling and use the screenedcandidate gene (HSP90, GPR23and ITG8)associated with heat stress for furtherfunctional verification.1.The candidate genes mRNA expression in different populations undercontinuoused heat stress: Each selected healthy Huainan ephedra chicken (H) andWenchang chicken (W)126roosters at8weeks of age as experimental animals,Increased the room temperature rapidly to (35±1)℃,what was as the starting point(0h), kept the temperature (35±1)℃for72h,then return the temperature to roomtemperature24±1℃. Choose the hypothalamus at0,12,24,72and recover to12,24,72h. Quantitative PCR were used to detect the HSP90, GPR23gene expression changingin the in different varieties.The Result shows that At0-24h, HSP90express at thesame trend under heat stress in the H and W hypothalamus. Rise and then decrease,reach the highest expression at24.And HSP90in H hypothalamus expressed higherthan W at each time point.The expression has continued to rise and h; W in24h torestore normal12h expressed decreased significantly (P <0.05), reach the highestexpression at24.Then it has decrease tend.2.SNPs detection.Through direct the DNA sequencing of candidate genes (HSP90, GPR23, ITGA8) SNP in some exon and intron regions, there has each mutation in the HSP90W4-87, GPR23A2-767G and ITGA8A4-2798G location, and HSP90W4-87was located in the fourth exon, but it doesn’t cause the amino acid changed. The other two SNPs are in GPR23sencond intron and ITGA8fourth intron.3mutations in theH and W are in Hardy-Weinberg equilibrium, but has no significant difference in theW and H (P>0.05).Conclusion: During the high temperaturet to normal temperature process,the expression of HSP90, GPR23in H and W in hypothalamus,change regularity, inferred thatthe HSP90, GPR23have a Relationship with heat stress control a, but the exact molecular mechanism need further study.
