Aseptic seeding and the establishment of regeneration system play an important role on the success ofcross-breeding of orchid, since these two factors determine the success of seed germination and help toobtain the object plant. It is necessary to have a further research on these two factors. This experimenttakes the capsules of Cymbidium hybrid as materials and its main purpose is to prove that the asepticseeding are influenced by the factors such as different pick times, seed pretreatment, basal medium,growth regulators, organic additives et al. The object materials of this dissertation are rhizomes whichgerminated from hybridization seeds of Cymbidium goeringii×Cymbidium hybridum, This dissertationsystematically studies factors which influence proliferation, differentiation and seedling rooting ofrhizomes during the growing period. The results are as follows:1. The best pick time of the capsules was6-8months after pollination.2%NaOH solution and0.5-5%NaClO solution were favorable to the pretreatment of hybrid seed germination; KC as the basalmedium and stationary culture were suitable for the germination of hybrid seeds. KT was better than6-BA for seed germination on total effect,6-BA1.0-2.0mg/L was better for seed germination;Activated carbon had the greatest influence on seed germination, followed by6-BA and NAA, coconutjuice had minimal impact for hybrid seed germination, coconut milk was not suitable.The bestcombination of various factors was that:NAA0.5mg/L+6-BA1.5mg/L+AC1.0g/L.2.1/2MS was the suitable basal medium for protocorms of filial generation.The effect of2,4-Dand KT on differentiation was not significant and6-BA1.0mg/L+NAA1.0mg/L was the bestcombination for proliferation and differentiation. Organic additive coconut milk had the most significantinfluence on the proliferation of protocorms, followed by banana mud and potato juice. Here100g/Lbanana mud and50ml/L coconut milk was favorable to the proliferation and differentiation of theprotocorms. Uncutting and1.0cm as cutting method and size was suitable for the proliferation of theprotocorms. TDZ0.1mg/L was suitable for the differentiation of the protocorms. The best combinationfor the differentiation of the protocorms was:1/2MS+NAA0.5-2.0mg/L+TDZ0.15mg/L+potatojuice90g/L.3. MS was the optimal medium for seedling root culture, followed by KC and1/2MS. GA30.2-0.5mg/Land NAA1.0-2.0mg/L were better for seedling roots. Organic additives potato juice60 g/L+banana mud60g/L worked best on the seedling roots. KC+NAA1.5mg/L+GA30.1-0.5mg/L+banana mud90g/L was better for hybrid rooting in the orthogonal combination.4. When transplanting, selecting seedling rootstocks of180d and smelting plants a week was better,The most combination for transplanting seedlings is two pine bark, one pearlite.
