Transcription factors (TFs) as one kind of important protein can bind to the cis-element and regulate the expression of target gene. Both the normal development and the proper response to environment of plant depend on TFs. WRKY TFs comprises a large gene family in higher plants. The wide distribution of WRKY genes indicates that this gene family may have an extensive role in plants’developmental and physiological processes. Since the identifica-tion of the first WRKY protein (SPF1) in sweet potatoes, numerous WRKY TFs were widely found in plants, such as74genes and102genes in Arabidopsis thaliana and rice (Oryza sativa), respectively,97genes in sunflower (Helianthus annuus),46genes in canola (Brassica napus L.) and197genes in soybean (Glycine max). The wide distribution of WRKY genes indicates that this gene family may have an extensive role in plants’developmental and physiological processes.WRKY6gene was cloned in potato, it’s function is not clear, so we did the following experiment to study it’s function, The processes and results can be summarized as follows:1) WRKY6gene’s pGBDKT7plasmid was constructed and was inducted into AH109type yeast, then transcription activation was been found in WRKY6gene.2) A specific fragment was choose from the WRKY6gene to construct RNA-interference plasmid for further research.3) Expression quantity of WRKY6gene in root, aboveground part and tuber show no obvious difference illustrated that WRKY6gene has no tissue expression difference.4) Abiotic stress test show that after treated with low concentration’s K+StWRKY6gene has the highest expression quantity in the3rd hours then reduce quickly. Low concentration’s phosphorous,10%PEG, low temperature and NaCl have no obvious influence in StWRKY6gene’s expression.5) Phenotype research of transgenic tobacco which was treated by low concentration’s K+, results show that WRKY6gene could response to K+. K+content test show transgenic line’s K+content higher than non-transgenic plant notably under normal water cultivation and low K+water cultivation.The preliminary studies prove that the StWRKY6transcription factors, were regulated by some abiotic stresses. The results laid the foundation for the further research on molecular mechanism of the regulation of gene expression, and provide a theoretical basis for the breeding of high-resistant plants.
