SiPEBP isolated from saussurea involucrata, is a cold regulation geneassociated with cold resistance. In this study, the binary expression vector of SiPEBP was builded, and which was transduced into the existing early-season rice parents as R996, ZR02,ZhulS, Xianling628S which were used to breed super hybrid rice in Yangtze river area. Research induction rate,and observe agronomic performance of transgenic lines under the artificial low temperature treatment. The main research results are as follows:1. In this study, as the super rice parent R996, ZR02, ZhulS, Xianling628S for receptor material, the target gene SiPEBP was successfully transduced into receptor R996and ZR02by agrobacterium-mediated method.2. Induced R996, ZR02, Zhu1S and Xianling628S by the MS medium with the same formula. After constant temperature induce for7days, induction rates of above four rice varieties were87%,84%,73%,82%, that showed the four varieties have different adaptability of the same formula MS medium.3. Dip R996, ZR02and Xianling628S callus with different level of Agrobacterium liquid, which found the most suitable for the three cultivars when ODgoo values in range of0.4-0.6, the effect is best.4. Callus of R996, ZR02and Xianling628S with different co-culture time training, after3days, all of the three varieties transient expression rate was the highest,3days is the optimal co-culture time.5. The PCR identification of transgenic offspring and genetically modified plants SiPEBP analysis indicated that hygromycin HPT marker gene and SiPEBP of saussurea involucrata both have been integrated into the genome of indica rice.6. Transgenic rice lines1-9,2-10and receptor R996on cold tolerance test:three material is placed in the Light incubator under15℃treatment for10days. The results show transgenic plants is more cold tolerance than receptor.7. Compared the yield of1-9,2-10, R996, the result shows that the exogenous gene have broken R996balance, between the transgenic and receptor yield difference. And from the same strain offspring materials also appeared separation.
