In this study, used different NaCL concentration treat three different varieties of potato plantlets (C3, C5, Atlantic). Determination of Potato Plantlets protection enzyme gene (SOD gene) expression changes and physiological and biochemical indexes of potato plantlets under different concentration of salt stress.Physiological and biochemical changes of Potato plantlets under different concentrations of salt stress (0.0%,0.2%,0.4%,0.6%,0.8%) were detected, growth of the plantlets less affected under0.2%concentration of salt stress,plantlets groundgrowth and underground growth more affected under0.4%concentration of salt stress, the underground part of the potato plantlets growth was significantly affected and growth of the ground parts is very slow under salt stress concentration of0.6%. the whole plant growth inhibited under0.8%concentration of salt stress.The physiological and biochemical indicators (chlorophyll content, average root length, fresh weight, proline content, SOD activity, POD activity) of three varieties of potato plantlets under salt stress has been detection.From the test results that salt stress concentration of between0.0%-0.2%, the indicators are declined slightly excepted the POD activity, and with the further increase of the concentration of salt stress, show a further downward trend. POD activity slight rise at0.0%-0.4%of the concentration of salt stress, a decrease in POD activity when the concentration of salt stress more than0.4%. excepted the POD activity, all indicators are reduced with the increase of the concentration of salt stress.Extraction of total RNA of potato plantlets under different concentrations of salt stress, reverse transcribed into cDNA after reverse transcriptase, RT-PCR detection found that the expression of SOD related the gene (PGSC0003DMT400070920) Withsalt stress concentration increased gradually decreased, showing a decreasing trend, under0.2%salt concentration treatment, the Potato Plantlets the ground and the underground part of the expression level decrease of29.9%and14.3%respectively.under0.4%salt concentration treatment, the Potato Plantlets the ground and the underground part of the expression level decrease of37.7%and26.5%respectively.0.6%salt treatment, the potato underground part of the expression of a decrease of35.3%.
