Sacbrood bee virus (SBV))is a wildly spread honey bee virus around the world. Thepathogen of sacbrood bee disease is a kind of linear single stranded RNA virus that with around-hexagon envelop from28~30nm diameter. Because western honey bees owns ahigher resistances to SBV, so they can cure themselves after infected by SBV. SBV won’tcourse severe commercial loses. For eastern honey bees, they were very sensitive to sacbroodvirus, so once the Chinese sacbrood bee virus(CSBV) infected Chinese bees, it becomepandemic, and coursed serious economic loss.In this study we investigated the occurrences of CSBV in loess plateau including Shaanxi,Ningxia, Shanxi, Gansu provinces from20th centuries. Also we invented a new identificationtechnique——gradient band PCR method and put it into firstly application. The mainlyresults in this research are as follows:(1) There are three pandemic of CSBV from1970s till nowadays in loess plateau. The firstpandemic was in early1970s, the second one was taken from late1980s to early1990s andlast one happened in these5to8years. CSBV coursed one of the most severe disease thatconstituted an obstacle to our native honey bee industry, they often causes enormous losses toswarms.(2) A novel method for identification of CSBV, Gradient band PCR (GBP) was set up inthis study. Compared with traditional RT-PCR, this new identification method was very quickand easily manipulated, which provided another method to diagnosis and control the Chinesehoneybee sacbrood. The healthy samples and CSBV infected bee larvae from south ofShaanxi, north of Shaanxi and middle of Shaanxi was confirmed by both of traditionalRT-PCR and gradient band PCR, respectively. The PCR products was observed by AGE andthen sent for sequencing. The results showed that the identification accuracy were the same byboth RT-PCR and GBP, and the GBP method was a very quick, convenient and comparativelyeasy approach that could identify the virus without the results of PCR product sequences.
