In this study, to discuss the differences of the grain weight and the total grain proteincaused by the dwarfing genes Rht10, we have studied the grain filling characteristics andproteomics of the tested wheat (Lumai15and Rht10lumai15). And the main results were asfollows.1.The survey of grain kernel weight and grain filling characteristics show that the grainkernel weight of Rht10lumai15less than Lumai15in the same period, after mature Rht10lumai15less than Lumai159.83%; the reason caused grain weight reduction is that Rht10reduced grain filling rate and shorten the duration of grouting.2. Dwarf gene Rht10in grain formation, filling and maturing on wheat grain protein mayhave an impact. In grain formation, Rht10lumai15were up-regulated7protein spots,down-regulated expression of8protein spots; in grain filling, Rht10lumai15wereup-regulated8protein spots, down-regulated expression of6protein spots; in grain maturing,Rht10lumai15were down-regulated expression of13protein spots.3. We have cleared the function of most identified proteins by comparing the results ofmass spectrometry and the amino acid rank in protein databank. And we have classified all theidentified proteins according to the function of the protein, mainly included proteins related tocarbohydrate metabolism, enzymes related to protein synthesis, enzymes associated withenergy metabolism, proteins related to material transport, proteins related to disease resistance,cold shock domain protein and serpins.4. The results show that fructan synthesis build is limited due to reduced grain volume,starch, glucose and glutenin synthesis decreased due to reduced dry matter accumulation,class glycolipid transfer protein and decreased expression of annexin cause limited masstransfer between the source and sink, accumulation of toxins cause cell toxicity, transcriptionand translation efficiency leading to cell dysfunction, these phenomena may make grainfilling rate and grain weight reduction reasons. Rht10through these pathways directly orindirectly affect the grain development. Through analyzing the grain filling characteristics and proteomics of the tested wheat,we have found that the dwarfing genes Rht10may inhibit or delay some proteins expression,which related to nutrients synthesis, transport, transformation.
