In the process of growth and development, wheat is often subject to disease、drought、high temperature、high salt and other biotic or abiotic stresses, seriously affect the quality and yield of wheat. AtNPR1、PPTPSI, OoHSP18I、ScPRX1four genes are all plant resistance related genes. In this study, wheat was transformed with Biolistic Bombardment and Agrobacterium tumefaciens carried these four genes in order to obtain transgenic lines. Therefore, it is significance for the breeding of resistant varieties of wheat.The gene of AtNPR1is plant disease-resistant gene, it is a signaling regulatory elements of SAR pathway and it plays an important role in regulating the occurrence of plant SAR. By Biolistic Bombardment and Agrobacterium-mediated two methods,AtNPR1gene was transformed to Ningmai13immature embryo.59transgenic plants were obtained,8positive plants were confirmed through PCR, the overall conversion rate was0.50%.7transgenic positive plants self-T1generation be used as seedlings stage phenotypic of wheat gibberellic disease resistance identification and field single flower infusion inoculation identification, The result showed that the transgenic plants T1generation compared to receptor have different degrees of resistance to improve.In adversity stimulus, the content of free proline(Pro) in plant increases significantly. Varieties with high ability of resistance accumulate more proline, Superoxide Dismutase(SOD) is an enzyme containing metal. The enzyme exists in aerobic organisms prevalently, helping cell to resist damage from superoxide anion free radical, also reflects the ability of resistance. Therefore Pro and SOD reflect the resistance of the plants as well as tolerance. Determination of the content of free proline and SOD in part of T1generation, the result showed that:after adversity stimulus, most of transgenic plants contain more proline and SOD’s activity than the control groups, the AtNPRl gene had expressed in a certain degree.PPTPSI gene is encoded physcomitrella patens trehalose synthase gene, can improve the drought resistance and cold tolerant of plant, the OoHSP18I gene is isolated from Oenococcus oeni. It is encoded HSP18protein, can improved the heat resistance of plant. The ScPRX1gene can encoded peroxiredoxin, increase the resistance of plants. In this study, Y14mature embryo was transformed with Agrobacterium tumefaciens carried these three genes.34transgenic regenerated plants were obtained,27positive plant were confirmed through PCR, in regenerated plants the proportion of positive transgenic plants is higher, the overall conversion rate was0.63%.
