Expressed genes in broilers spleen and bursa are potentially associatedwith the body’s immune genes, we study19expressed genes (known and unknown EST) ofspleen and bursa which got from our team to study its expression features further and tostudy the30%energy restriction on broiler spleen and bursa candidate gene expression, Inthis study, mainly carried out from the following two parts.Test oneIn order to detect these candidate gene and sequences expression differentially invarious tissues, we choose7-day-old Dwarf Silky Silky as the object in this study, and usingsemi-quantitative PCR detect relative expression levels of these candidate gene andsequences in the muscular stomach, intestinal, glandular stomach, brain, liver, breast muscle,cerebellum, legs, heart, lung.Test two4018-days-old Arbor Acres female broilers were used to investigate the30%energyrestriction on broiler spleen and bursa candidate gene expression. The chickens wererandomly allotted2treatments(20in each group),each bird raised in one cage for30days.Then slaughter, gathering the spleen and bursa tissues, energy limit group and freedom togather the food group each20samples, repeating units for a single individual. According tothe candidate genes and related sequences, using fluorescence quantitative RT-PCR carrieson the quantitative analysis of candidate genes and related series. Following was the results:(1) In the spleen tissues, in which7differentially expressed fragment showed aupward trend in energy limit group, a downward trend for10, while DAD1and ZNF644showing significant differential expression (p <0.05), DAD1showed a downward trend,ZNF644showed a upward trend.(2) In the bursal tissue, in which5differentially expressed fragment showed a upwardtrend in energy limit group, a downward trend for12, while ACTG expression significantdifference (p <0.05) and showed a upward trend.(3) Through bioinformatics analysis we showed that: DAD1and ZNF644areassociated with the disease; ACTG is the highly conserved genes which exists in most celltypes.In this study, We identified differences in energy restricted broiler spleen and bursa displayed EST through quantitative analysis. This provided important information forfurther study of energy restriction on broiler immune organs, and provided a reference forhuman researching energy restriction effects in broiler from the molecular level.
