Chinese fir (Cunninghamia lanceolata (Lanb.) Hook.)is one of superior tree species which is economic benefit remarkable and high ecological benefits. In recent years, by selective breeding, there are a large number of excellent clones of Chinese fir in our country, and some of the clones has been established rapid propagation system, the related clones have achieved the technique of industrialized growing seedling, which made great contribution for the development of plantations. It has not yet been carried out in southwest of China, Chinese fir seed micro-propagation, because of the different Chinese fir clones, its genetic traits have bigger difference, thus in the process of tissue culture proliferation rate also has a big difference. This difference has greatly influenced Chinese fir clones fine soma-clone of popularization and application. Therefore, through two years of experiments, this paper established a relatively stable fir in vitro rapid propagation system, completed the fir test-tube seedling root technology exploration of the bottle, and use the Chinese fir young stem section for embryonic callus induction.The main results are as follows:1. The best disinfection scheme of Chinese fir’s young stem:Mild detergent water or detergent water soak for30min, rinse under running water for3-5h,0.1%carbendazim soak for30min, cleaned and placed in the laminar flow cabinet, then disinfected by a20s immersion in70%ethanol foolowed by5min immersion in0.1%(w/v)mercuric chloride solution and then rinsed five times with sterile distilled water.2. The best time for collecting the shoot segment explants of Chinese fir is in late march, which is early spring in the place. 3. For maintenance and proliferation of Chinese fir terminal buds, the best media is MS containing0.4mg/L6-BA,0.5mg/L IBA,30g/L sucrose,0.7g/L agar; For maintenance and proliferation of fir shoot stem segments, the best media is MS containing0.7mg/L6-BA,0.2mg/L IBA,30g/L sucrose,0.7g/L agar; The best media for tissue culture seedling rooting is1/2-1/4MS(basal salts were reduced at half to quarter concentration from the standard) containing0.4mg/L6-BA,0.5mg/L IBA,30g/L sucrose,0.7g/L agar.4. The best treatment for Chinese fir tissue culture seedling rooting outside of the flasks:the volume ratio of loess to perlite to vermiculite is3:3:1, remove part of the callus and50mg/LABT1#, Air humidity85-90%, temperature of20℃, low light.5. The in vitro seeding of transplanting:the volume ratio of loess to perlite to vermiculite is3:3:1, with1/20MS (basal salts were reduced at1/20concentration from the standard) solution as a nutrient liquid and conduct foliar fertilizer to the seedlings, then the survival rate of transplanting could reached70%.
