In animal liver cells, lipodystrophy will cause the abnormal deposition of fat, thus result in fatty degeneration of liver. This is due to the change of the expression quantity of some lipid metabolism factors,which are affected by nutrients. The Intrahepatic lipid deposition and the lipoprotein secretion balance destruction may be the main cause of hepatic steatosis. VLDL is a way of secreting TG and TG are assembled in the liver cells into VLDL-TG,which are secreted out of the cell wall. These series of effections are closely associated with glucose.In this study, with Sichuan white goose for experimental object, The modified semi-situ "Seglen two-step collagenase perfusion" method was used to separate and collect the goose hepatocytes, and to be cultured with glucose in vitro. We measuredthe effect of different concentrations of glucose on the function and bioactivity of hepatocytes, and also detected the effect of different concentrations of glucose on the VLDL concentration and the mRNA expression level of MTTP and ApoB genes.The main resultsare as follows:(1) The cells activity increased significantly with glucose concentration increasing, which is most visible in a high dose glucose (35mmon/L), and it reached56.67%;(2) When goose hepatocytes were incubated at the same glucose concentration, with the increase of culture time, the hepatocytes activity increased.(3) The TG and TC concentration increased with the increasing of glucose concentration cultured with goose hepatocytes.(4) Detection the ALT and AST levels can response hepatocytes damage, and the results found that within a certain range of glucose concentration (25mmol/L), hepatocytes damage was not obvious, and when hepatocytes were cultured with high glucose (35mmol/L), cell damage was obviously (P<0.05);(5) The TP concentration slightly increased with the increase of glucose concentration, while ALB concentration significantly decreased, which indicates that the cellular protein synthesis capacity reduced with the increase of glucose concentration.(6) In goose hepatocyte, low concentration glucose (5mmol/L) did not significantly impact on the intercellular concentration of VLDL, the intercellular concentration of VLDL significantly improved (P<0.01) under15mmol/L glucose while25mmol/L and35mmol/L glucose could significantly improve intercellular concentration of VLDL (P<0.001);(7) MTTP mRNA expression was inhibited after the culture with glucose, under the effect of low glucose (5mmol/L) and high glucose (30mmol/L), MTTP expression levels were reduced by13%and26%, respectively.(8) Glucose cloud increase the expression of ApoB gene, and with increase of glucose, the mRNA level of ApoB increased. Compared with control group, under the effect of low glucose (5mmol/L) and high glucose (30mmol/L), MTTP expression levels were increased by52%and78%, respectively.
