SOC1(SUPPRESSOR OF OVEREXPRESSION OF CO1)/AGL20(AGAMOUS-LIKE20), onemember of the MADS-box family, plays an important role in regulating the flowering of the plant,it can integrate the signals involved in photoperiod, vernalization, gibberellin and autonomouspathways, interact with many other genes to regulate the flowering time, floral patterning andfloral meristem determinancy. The identification of SOC1is important to the research offlowering mechanismn in bamboo, a group of perennial evergreens with unpredictable,long-periodic, gregarious, and uncontrollable flowering time. BoSOC1and PvSOC1were clonedfrom the first strand of Bambusa oldhamii and Phyllostachys violascens cDNA through RACEusing primers designed according to the homologus genes conserved region of Oryza sativa,Triticum aestivum, Zea mays, Sorghum bicolor and the transcriptome database of Phyllostachysviolascens. Five different sequences of BoSOC1, named BoSOC11-3, BoSOC11-5, BoSOC1Q-2,BoSOC1Q-8, BoSOC12-5, are obtained. The BoSOC11-3’s ORF is666bp and codes221aminoacids; BoSOC11-5’s ORF is669bp and codes222amino acids; BoSOC1Q-2’s ORF is672bp andcodes223amino acids; BoSOC1Q-8’s ORF is669bp and codes222amino acids; BoSOC12-5’sORF is675bp and codes224amino acids. One PvSOC1sequence is obtained with an ORF of681bp coding226amino acids.Homology analysis indicated that the BoSOC1protein was highly homologous to other SOC1proteins from grass family, such as Oryza sativa, Triticum aestivum, Zea mays, Sorghum bicolor,and could gather into one group with them. RT-PCR analysis shows that transcripts of BoSOC1inflowering buds is about12fold greater than in vegetative shoots, and transcripts of PvSOC1inmature leaf is much more than other aboveground parts of flowering Phyllostachys violascens andvegetative bud’s is the least. However, the transcript of PvSOC1in vegetative bamboo is thevegetative bud’s highest and the stem’s least.35S::PvSOC1Arabidopsis plants have less rosettleleaf numbers and display early-flowering phenotypes accompanied with apparent wrinkle andexstrophy of petal. The leaf of transgenic Arabidopsis is wider, shorter and lower brighten than thewild type. These suggest that PvSOC1promotes flowering during plant growth, and may play rolein other physiological activity.
