Wild cannabis germplasms is not only the natural gene bank of cannabis researchand utilization, but also the important basis of hemp breeding. In this article, geneticdiversity of12wild cannabis resources and4cultivars were assessed by the methods ofphenotypic characters and RAPD markers, and the cluster of phenotypic traits and RAPDMarkers were constructed by Farthest-neighbor method and UPGMA method respectively.2wild cannabis resources and2cultivars were used as materials to study the effects ofdifferent water stress on physiological indexes of cucumber seedlings,and to providetheoretical basis for research on drought-resistance mechanism and cultivation ofcannabis. Recorded the variations,The four kinds of marijuana leaf and plant height indrought, Drought treatment were measured in different periods the chlorophyll and MDAcontent in leaf. The following is the main results:1. the genetic diversity in wild cannabis was abundant, and the difference of11Morphological characters in all germplasms was significant (P<0.001). Thethousand-grain weight had highest coefficient of variation2. Based on the concentration of PCR reaction process and system factor comparisontest, The result was to set up the reaction system of RAPD, RAPD reaction system:DNA(40ng),rimer(1.0umol/L),Mg2+(1.5mmol/L),Taqpolymerase(2.5U),dNTPs(0.2mmol/L).3. Ten primers of14base pair were screened out from70random primers and wereused to analyze with RAPD among16accessions, which generated106bands, ofwhich polymorphic bands number was74.52%.4. The whole germplasms could be classified into three groups based onMorphological cluster. The first group was composed of12wild germplasms which further divided into two subgroups based on the degree of latitude, while The othertwo groups contain only3cultivars.5. RAPD cluster analysis showed that the whole germplasms were also classified into3groups clustered with the characteristic of regionalism. Germplasms in Yunnan andXinjiang were in one group, Germplasms in northeast and Huabei were in the secondgroup, while Germplasm in Xizang was alone in another cluster, the wild andcultivars were not distinguished obviously.6. Based on experiment of p ot-cultivating under different degree of drought stress for4hemp cuhivars:M50,M448,Yunma1,Yunma5,their drought tolerance,includingphysiological characteristics and growth, were comprehensively evaluated andcompared. The results showed that with the increase of water stress, Plant height isrestrained, the content of chlorophyll decreased, MDA content along with the increaseof water stress degree is obviously rising trend.The results suggest that the genetic diversity of cannabis in china is complicated,Comprehensive analysis combined with phenotypic and genetic loci.
