Along with the gradual completion of rice genome sequencing, gene pool is ceaseless and perfect, rice has become a model organisms in the study of monocotyledonous plants and grasses. In recent years, many important gene related to drought resistance, florescence, fertility and other agronomic traits was successfully cloned. Rice florescence, also known as the heading stage, vegetative to reproductive growth is a crucial transition. So clone the genes that control rice blossom far-reaching significance to study rice florescence.It can help to improve the regulation of blossom gene system.At the same time, the breeding work will be guided in genetics.Through the expression technology (OverExpression) and RNA interference (RNAi),we can clone rice blossom related genes:CDKl, CDK2, CDK3, CDK4, CDK5. First of all, cDNA gene sequences was amplified by RT-PCR technique.Then designing the OE primers and RNAi primers and cloning full length fragments and interference fragments of CDK1, CDK2, CDK3, CDK4, CDK5genes with cDNA template by using technology of PCR, Connect the OE fragments to pCAMBIA1300-NOS-35S expression vectors,connect interference fragment to pCAMBIA1300through subcloning technique, Finally get10construction completed cloning vectorThe last fragment can be transferred into Agrobacterium to make transgenic experiment step. The gene cloning can provide materials for transgenic experiment at next step, also make a foundation for the study of gene function and plant traits.
