Popular(Populus simonii×Popular nigra) is one of the important industrial and economic tree species, is a model plant for forest research. It is found that the juvenile period of popular is longer, whereas many traits only express when adult. Therefore, stimulate precocious flowering and shorten breeding period become the hotspot of research. In recent years, successful of transfermation of many genes associated with flower development, laying foundation for plant flowering stage modification through transgenic technology. Many studies about API transgenic Arabidopsis and Antirrhinum showed early flowering ability, which reported rarely in poplar. And the researches about transgenic poplar are most based on heterozygous diploid.The previous master of our obtains haploid poplar "B6"which is derived from female Populusxxiaohei by natural pollination by another culture. Chromosome of somatic cells of haploid Populusxxiaohei will be spontaneous doubling during the subculture, and formate homozygous diploid which is called double haploid. For long-time observation, we found the appearances of double haploid poplar and diploid poplar were different. The purpose of this paper is to get the positive plants of double haploid poplar and diploid poplar for API gene. Using Solexa sequencing technology, we build four DEG libraries for leaves and shoot tips of double haploid of poplar and diploid of poplar, then analysis the difference gene expression of the four libraries. The XVE inducible system with GUS gene was constructed, and determined the best suitable concentration of estrogen which inducibled the expression of GUS. Results obtained from our experiments as following:1.2x35S::APl pB2SM,35S::GUS PER10vectors were constructed, obtained the positive plants. The positive plants carried out phenotype observation, double haploid poplar showed early flowering and after15days, the transgenic plants began to wither then death. But diploid poplar did not have that appearance.2. Analysed the different gene expression profile between the control plants of double haploid of poplar and diploid of poplar using Solexa sequencing technology. In these different genes, we found the genes on Photosynthesis, synthesis of plant hormones and the development of phloem and xylem, which expressed down regulation in double haploid poplar.3. The best suitable concentration of estrogen which inducibled the expression of PER10-GUS in double haploid of poplar was5umol/L.
