The yield and quality of crops are infected by the impact of plant diseases, the safety issueshighlighted in the agro-food production and people ’s living suffer a serious hazard.Fusariumgraminearum as a semi-vivo nutritional fungus can infect a variety of food and cashcrops,such as barley, etc.,can cause great harm and loss.Arabidopsis-Fusarium graminearum interaction model for us to effectively provide a modelsystem to study the interaction between plant pathogens. EDR3(enhanced desease resistance)is one of Arabidopsis powdery mildew resistance gene. edr3that enhance cell death can beinduce Arabidopsis plants resistant to powdery mildew, edr3mediated Arabidopsis resistancedepends on the salicylic acid signaling pathway, rather than dependent on jasmonic acid andethylene signaling pathway. We do not know how EDR3regulation the reaction ofArabidopsis anti-scab.Because edr3-mediated resistance play which role in which signaling pathway is not yetclearly.We selected genes which occupy a prominent position and have representative in theSA/JA/ET signaling pathway with edr3to be the double mutant hybrid materials usingbiological methods to obtain.Identification of the pure double mutant edr3/ein2,edr3/coi1,edr3/sid2,edr3/pad4,edr3/nim1.To study the functional of EDR3.Fusarium graminearum is a non-host resistence semi-biotrophic fungus for Arabidopsis,which can infect a variety of cereal crops,including wheat and barley,causing Fusarium HeadBlight (FHB).Because of the large size of wheat genome,long life cycle of wheat,theinteraction between the F.graminearum and its hosts is still unclear.In this study,because F.graminearum infect model plant Arabidopsis thaliana,so we used it as a model system tostudy how plant defends the infection of F.graminearum. Mutant edr3,which was identifiedpreviously to mediate with different pathogens, was used in this study.Experimental resultsshow that the enol (DON) bacteria without deoxynivalenol sickle F.graminearum sporesuspension was puncture infect Col-0and edr3,observed8days after inoculation,the bacteriacan successfully infect its non-host plant Col-0and edr3.Surprisingly,we found that mutant edr3was more sensitive than wild type Col-0to theinfection of F.graminearum.Furthermore,we also infected double mutants edr3/ein2,edr3/coi1, edr3/sid2, edr3/pad4by F.graminearum,and found that the susceptibility to F.-graminearum mediated by edr3was dependent on ET/JA signaling.These results suggest thatEDR3plays a positive role in the resistance to F.graminearum, which was help to agriculturalproduction. |
