Mannan-binding lectin is an important component of animals’natural immune system, it plays an significant role in resistance to external pathogenic microorganism infection, MBL can activate the complement system to carry immunity regulation function.Bovine MBL1was studied in the experiment as following:First of all, the Holstein bovine MBL1level in serum was detected through the enzyme-linked immuunosorbent assay(ELISA); secondly to extract the whole DNA in blood, MBL1gene promoter region was amplified by PCR, then using of methylation sensitive restriction enzyme to digest the target fragment; Finally, bovine MBL1-CRD was cloned and determined using RT-PCR technology.The MBL1level of serum is between12.3~109.69μg/L through the ELISA detection, it is consistent with the reported level. Bovine blood DNA was extracted by ELS Pyrolysis, and a pair of primers were designed to amplify MBL1gene promoter region fragments, the promoter region was digested by Hha I, the result indicated that two fragments were found to proved there have no methylation of enzyme loci. There is385bp in MBL1-CRD region, it can translate122amino acid residue. By sequence alignment,there is68.15%homoloy between bovine MBL1-CRD region and porcine MBL1-CRD region at nucleotide level, the homology between bovine MBL1-CRD and sheep MBL1-CRD was93.77%, it showed CRD region among bovine and porcine、 sheep has high homology.This study on the bovine serum MBL-1level detection, has important biological significances to study correlation between MBL1with infectious diseases. In the meanwhile, the exploration of promoter region methylation, and the genetic analysis for the MBL1gene CRD region provide a basis for research the function of bovine MBL1.
