Four Ccucurbita maxima lines were used to construct3six generation populations to investigate the genetic rules of fruit color, length of internode, length of seed and width of seed.1156SSR primes were used to construct a linkage map and locate the fruit color in the linkage map initially.This research will provide supplement for the breeding in fruit color, bush type selection and seed size selection. The SSR genetic map will give a acceleration for MAS, and provide a supplement for molecular clone in cucurbita maxima further.(1)A yellow and a green fruit color habit accessions and their reciprocal Fis and F2S of Cucurbita maxima were compared for fruit color. The result show that fruit color is controled by cell nucleus, full green is recessive to not full green.(2)Bush and vine habit accessions and their reciprocal Fis and F2S of Cucurbita maxima were compared for internode length. The result showed that developmental reversal of dominance were found in this experiment. When plant were young the internode length were controled by two gene and by single gene during developmental reversal of dominance, finally controled by single gene and poly-gene, environment is very important after the developmental reversal of dominance. In1-5and in6-15internodes the genetic model fit the El model,16-25internodes the genetic model fit the D4model both.(3) A big seed and small seed habit accessions and their reciprocal Fis and F2S of Cucurbita maxima were compared for length of seed and width of seed.the result show that the length and width of seed are all control by cell nucleus but not cytoplasm genome.Length of seed and width of seed are all fit the D2model, and the main gene have a strong effect to trait, can explain more than75%of the genetic variance. The corelation analysis of length and width show that there are a significant corelationship between length and width,and the corelation coefficient can reach0.99.(4)178SSR primers were polymorphic between two parent in1156SSR primer.171F2were used to construct the first SSR linkage map. Finally,15linkage groups were constructed and contain98loci. The full length of the linkage groups reach1549.3cM and15.8cM between every loci, gf were located in LGm7, and the most near loci is PU000529, the linkage distance is6.3cM.
