Avain influenza virus (AIV) can lead to acute infection diseases, and is one of the most seriousdiseases threating the poultry industry in China. AIVs could cause disease outbreak in birds, and aredeadly pathogens in mammals too. Currently, our country AIV surveillance plays an important role incontrolling HPAIV subtypes. But, during the surveillance, we also isolated numbers of H6subtype AIVin China. This study sequenced surface genes of92H6subtype AIV isolates from2010-2012, Ianalyzed18of them to investigate the antigenicity, pathogenicity and genetic evolutionary relationship.The result of analsis of18H6subtype AIVs revealed that they divided into4subgroups, and withsignificant changes in the group4among18tested viruses.The surface genes sequencing result showsthat N2/N6are the main subtypes of neuraminidase, and N2NA subtype is gradually replaced by N6.The whole-genome-sequenced virues showed the internal gene of18viruse are mainly from H6lineages, very few of them are H9/H5-like. Genotyping demonstrated that H6subtye virues have8genotypes, and all of them are reasserted. One genotype, represented byA/Chicken/Hunan/S1103/2010(H6N6), predominated in China during2010-2012,10out of18viruesare this type. Experimental infection tests showed that the isolates were able to replicate in lung, while15of them able to infecte shall. But none of them could replicate in other organs, and cause no death orsignificant weight change.This study use concentrated virus particles as immunogen, to immune5-6weeks old BALB/cmices.8positive anti-H6HA hybridoma cell strains were selected by hemagglutination inhibitionmethod. The specificity experiment showed:8McAbs could react with H6hemagglutinin trimer andcould cover84%of all the isolated H6subtype virues.These result sugessed that these McAbs would beused for study H6HA structure and function and develop rapid detection of H6Subtype AIVs.
