| This experiment used5different varieties as experimental material,studied the impactfactors of culture of in vitro female nucleus and plant regeneration.Mainly discussed theinfluence of embryoid culture from plant genotypeã€concentration of TDZã€time of heatshocking〠cultivation season,and screened appropriate culture conditions of embryoidculture.And preliminarily explored the impact factors of transplantation survival rate fromplant morphologyã€acclimation temperature and the way of hardening-seedling to acquire theculture system which is favorable for culture of in vitro female nucleus.The results of this experiment show that:1.Genotype has has a major influence on embryoid induction of culture of in vitro femalenucleus,there are relative difference between the time of embryoid occurring and embryoidinductivity.In the five materials,the embryoid inductivity of XH11-24is highest,and its timeof embryoid occurring is shortest, respectively are18.90%and33d.2.Adding TDZ hormone to culture medium has a certain acceleration on embryoidinduction.In the five concentrations, adding0.06mg/L TDZ to medium,the inductive effect ismore better,the embryoid inductivity is highest,is17.17%.3.Under the same heat-shock temperature(35℃),different heat-shock time has differentinfluence on embryoid induction.In the five time,120h has most ovules turning green andhighest embryoid inductivity,respectively are86.76%and18.57%.4.In different seasons,the embryoid inductivity is different.X11-24and X11-55,theirembryoid inductivity of autumn materials are higher than spring materials,respectively above5.40%and3.43%.5.There is obvious difference on survival rate of transplantation between differentmorphology of regenerated plantlets.When they have5-6true leaves,the survival rate oftransplantation is highest,is87.61%.6.The survival rate of transplantation of regenerated plantlets have difference when theacclimation temperature is different.In the three temperature gradient, when acclimationtemperature is day temperature21℃-27℃ã€night temperature10℃-20℃,the survival rate oftransplantation is highest, is88.27%. 7.Different ways of hardening-seedling have different influences on survival rate oftransplantation.In the three different ways,firstly take illumination training and then takehumidity training,the survival rate of transplantation is highest,is90.47%. |
