| Take-all is a important infection of the root disease of wheat in the world, primarilycaused by Gaeumannomyces graminis var.tritici (Ggt). Efficient disease-resistant wheat arethe fundamental method of control of take-all disease. This study optimized evaluationsystem to the disease, but also increase the identification of different transgenic wheat andsynthetic wheat, focused on the effect of growth and development of transgenic wheat bypathogen,and the inhibition of the growth of pathogenic on resistant wheat. The results are asfollows:1. Principal component, correlation and analysis of significant analysed in identificationof the conventional wheat materials discovered severity, black stem area, yellow leaf rate andblack stem rate had significant or extremely significant correlation relation with otherindicators. The principal component extraction revealed information of four indicators weremore than85%, load coefficient are:0.628,0.827,0.723,0.802. Differences between theabove four indicators can reflect different resistant to take-all more comprehensive and moreaccurate.2. According to above indicators, the identification results showed that19materialsshowed better resistance in the586conventional materials, Wan44resistance level closed tothe high resistance, and other15materials’ resistance level reached in middle resistance;found11high resistance materials and14middle resistance materials in transgenic wheats, ofwhich5materials showed significant resistance during the whole growth period in fieldnurseries experiment; identification in artificial synthetic wheats got2high resistancematerials which were CI107and CI161, and11middle resistant materials.3. PCR analysis of TiERF1-RC7and MYB genes in wheat, found both two genes wereintegrated into the receptor Yangmai18, which positive rate reached100%, MYB transgenicpositive rate was87.5%. Combined with the resistant identification result, resistance totake-all of TiERF1-RC7transgenic wheat was more stable. 4. The experiment of soluble protein screened19transgenic wheat materials, which hadhigher content of soluble protein, including10materials whose protein concent reached15mg/g, and the protein content of receptor were in7.5~8.8mg/g, slightly lower than theuninoculated receptor wheats. The trendency of concent showed that the resistance andprotein concent were positive correnlation.5. Electron microscope showed that quantity of mycelium in10ZGY-92transgenicwheat were more and stouter, and whose infection ability also higher. On the contrary,mycelium was smaller, thiner, and infection ability lower on the root of Yangmai18. It wassuggested that the expression of genes may affect the growth and development of mycelium. |
