Salvia miltiorrhiza Bunge is a kind of dicotyledomous plants in the mint family which isused as traditional Chinese medicine with its dried root and rhizome (named Salvia). Inclinical treatments, Salvia is usually used for healing irregular menstruation, coronary heartdisease, angina pectoris, ischemic cerebral vascular and so on. But in recent years, with thedeterioration of the ecological environment, serious drought in local scope happens frequently.Studies show that plants under drought stress can inhibite plant growth and development andlead to changes in morphlolgy, physiological and biochemical aspects, and even death.Therefore, study on drought gene is carried out with a view to improve crop,tolerance todrought, to ensure stable production of crops.In this study, amplified fragment length polymorphism technology (AFLP) wasemployed again based on previous work of our laboratory and two molecular markers thattightly linked with drought gene were found. The main results and conclusions were listed asfollows:1By optimizing the traditional CTAB method, an improved pprotocol for high-quality DNAisolation was established. This protocol was simple and fast, which could efficientlyremove polyphenols and polysaccharides, and can be used in AFLP analysis directly;2A total of170primer combinations were screened between drought-treated and controlledfertile S. miltiorrhiza bulks.14primer combinations out of them showed stable differencewith the rate8.2%. Then these14primer combinations were subjected to population testincluding43investigated individuals. Only one primer combination E9/M3was foundtightly linked to drought gene and the recombination rate was6.98%. The genetic distancebetween this marker and drought gene was7.02cM. Sequence analysis result showed thismarker sequence contained246bp and had high homology with Arabidopsis and Oryzasativa Japonica genome.3Total384primer combinations were screened between drought-treated and controlledsterile S. miltiorrhiza bulks.15primer combinations out of them showed stable differencewith the rate5.0%. The15primer combinations were then subjected to population testincluding43plants. Only one primer combination E2/M5was tightly associated withdrought gene with the recombination rate4.65%. The genetic distance between E2/M5 marker and drought gene was4.66cM. The sequence analysis result showed that itconsisted of357bp and had high homology with Arabidopsis and Oryza sativa Japonicagenome.
