Wheat powdery mildew is a global epiphyte disease caused by Blumeria graminis f.sp. tritici. The wheat powdery mildew is one of wheat main diseases and causes seriously damage in China. Transcription factors are key proteins for regulating gene expressions. To study molecular mechanisms of transcription factors underlying growth, development, and pathogenicity of B. graminis will promote researches to find novel targets of chemical control and provide new sight and good idea for designing comprehensive management of Blumeria graminis. Prediction of a protein function is an important part in Bioinformatics. Prediction of transcription factors of B.graminis by bioinformatics methods can raise experimental efficiency, so bioinformatics is important in B.graminis’study. This study mainly included the following works:1. This thesis put forward a new method for screening transcription factors’and activation domains of B. graminis. Basic principle of the method was to clone the B. gaminis cDNA library to the DNA-binding domain of GAL4gene, and then transfered to yeast cells. Finally, activities of transcription factors were measured by the expression of reporter genes. Our experimental results show that the method was feasible and had some advantages, such as easy, simple,, highly sensitive, and so on. It had found that the activation domains of transcription factors of B. graminis had low homology and most of them were the acidic domain by bioinformatics analysis.2. We constructed a model for prediction of transcription factors’ activation domain of B.graminis prediction. The prediction of the model was based on grouped weights as features extracting method and weigh K nearest neighbor algorithm as classification algorithm. We verified the model by comparing the prediction data with our experimental results. The experimental results showed that the prediction model had great accuracy and efficiency.
