| Alzheimer’s disease,AD,called senile dementia additionally.It is one of the common age-related diseases taking place frequently. That is the treatment of AD which has been verified and get recognized,to achieve therapeutic effect by the AChE (acetylcholinesterase) inhibition,AChE inhibitors.The secondary metabolites of Huperzia serrata, Huperzine A,is one of the special efficiency drugs for the AD treatment. Compared with other drugs, it’s the advantages, significant effect, short cycle of action,little toxic and side effect and not easy to caused resistance to drugs. However, the growth rate of Huperzia is very slow at the same time, on the other side, there are different numbers and types of endophytes in its body. It’s so difficult to obtain large numbers of huperzia whether planted or tissue culture. Synthetic class of drugs is not only more expensive, but also more side effects. Therefor, the natural production of AChE inhibitors has become a very potential and value of research.We isolate purify and species identify183endophytic fungi from Huperzia (Huperzia Bernh.) in this article,these fungi are combined the same strain, and we establish the Huperzia endophytes library. Then through the experiments of AChE inhibition activity, we identify the strains which is activated of AChE inhibitory. ESSZ05601is the a fungus of Urocystis sp. Ustilaginales Tilletiaceae. During the detection of40different strains of endophytic fungi, the inhibition ratio of AChE about ESSZ05601is the highest,60.95%,and this is higher than Hup A,42.39%. At the same time, we also carried out HPLC (High Performance Liquid Chromatography) testing with Hup A, Hup B and Hup C as testing standard,and find out the same or closer species with the standard peak time. The results showed that the peak time of ESSZ05601is completely different with the standard type, but only a single peak in the the310nm UV absorption wavelength, and the peak area is very large. This indicates that this alkaloids is not Hup A,Hup B or Hup C, but this secondary metabolites is very likely to become an effective drug for the treatment of AD. |
